摘要:
研究凝血酶敏感蛋白1(TSP1)及受体CD36、CD47对人肝癌细胞株HCCLM3生长及细胞周期的作用,初步探讨TSP1可能的作用途径。方法:根据处理因素将细胞株分为:TSP1组(加入TSP1蛋白,40mg/L)、空白对照组、CD36阻断组、CD47阻断组。阻断组细胞接种后先加入5mg/L对应的单抗,待单抗与细胞充分结合后再加入40mg/LTSP1。用四唑盐比色法(MTT)、流式细胞术和ELISA法分别观察TSP1及CD36、CD47单抗对细胞生长、细胞周期、增殖指数(PI)及TGFβ1分泌的影响。结果:TSP1组、CD36阻断组、CD47阻断组细胞生长受抑,且CD36阻断组的细胞生长抑制率低于CD47阻断组。TSP1组G0/G1期细胞数增加,S期和G2/M期细胞数减少,与对照组和CD36阻断组相比差异有统计学意义。对照组、TSP1组、CD36阻断组及CD47阻断组PI分别为:(42.70±2.09)%、(27.85±0.71)%、(38.04±1.35)%和(31.78±0.43)%,各组之间两两比较,差异均有统计学意义(P均<0.05)。各组TGFβ1的含量差异无统计学意义。结论:TSP1主要通过与受体CD36的结合对人肝癌细胞株HCCLM3的生长增殖有抑制作用。TSP1对HCCLM3的抑制可能不是通过调节TGFβ1的分泌而发挥作用。
Abstract:
To explore the role of thrombospondin1(TSP1) on the growth and cell cycle of human hepatic carcinoma cell line HCCLM3,and study the effect of its receptor CD36 and CD47.Methods:The cell line was divided into 4 groups: control group, TSP1 group, CD36blocked group, CD47blocked group.The cell growth of the 4 groups were observed by MTT test,cell cycle and proliferation index (PI), by flow cytometry,and the secretion of TGFβ1, by ELISA.Results: MTT test demonstrated that the growth of HCCLM3 in each of TSP1 group;CD36blocked group and CD47blocked group were significantly slower than that in control group. Inhibitory effect of growth in CD36blocked group was lower than that in CD47blocked group. Flow cytometry for cell cycle indicated that the cell number in G0/G1 phase increased,and the cell number in S and G2/M phase reduced in TSP1 group compared with that in control group and CD36blocked group.PI in the above 4 groups was(42.70±2.09)%,(27.85±0.71)%,(38.04±1.35)% and (31.78±0.43)%,and the differences among 4 groups were significant. The average value of TGFβ1 in each group had no significant difference among 4 groups.Conclusion: TSP1 can inhibit the growth of HCCLM3 cell through combining with the receptor CD36,and blocking the cell cycle on G0/G1 phase. The effect of receptor CD47 was weaker. The inhibit effect may be not related to the synthesis of TGFβ1.
