摘要:
研究人脑胶质瘤组织中丝裂原激活蛋白激酶(MAPK)的表达及活化情况。方法:应用免疫组织化学技术观察新鲜冻存的16例脑胶质瘤组织标本(其中Ⅱ级9例,Ⅲ级7例)和5例脑外伤内减压术后正常脑组织中MAPK及磷酸化MAPK(pMAPK)的表达情况。结果:MAPK阳性表达位于胞质;pMAPK位于胞质和胞核,且胞核阳性信号较强。胶质瘤Ⅱ级、Ⅲ级和正常脑组织中MAPK阳性区域面积百分率分别为(37.78±4.05)%、(53.58±1.60)%和(23.12±2.43)%,pMAPK阳性区域面积百分率分别为(36.91±3.85)%、(53.14±3.47)%和(18.61±3.30)%。胶质瘤组织中MAPK及pMAPK阳性区域面积百分率均高于正常脑组织(P<0.05);胶质瘤Ⅲ级者高于Ⅱ级者(P<0.05)。结论:MAPK及其异常活化与人脑胶质瘤的发生发展有关。
Abstract:
To explore the expression and abnormal activation of mitogenactivated protein kinase (MAPK) in the development and progression of cerebral gliomas.Methods:Immunohistochemistry was applied to detect expression of MAPK and phosphorylated MAPK(pMAPK) in 16 cases of gliomas and 5 cases of normal control cerebral tissues acquired from decompression surgery after traumatic brain injury.Results: MAPK expressed in cytoplasm, while the expression of pMAPK located at cell nucleus and cytoplasm, especially in nucleus. In grade Ⅱ, grade Ⅲ gliomas and normal brain tissues, the percentage of MAPK expression was (37.78±4.05)%, (53.58±1.60)% and (23.12±2.43)% respectively; the expression of pMAPK was (36.91±3.85)%, (53.14±3.47)% and (18.61±3.30)% respectively. Expression of MAPK and pMAPK in gliomas was significantly higher than that in control tissues(P<0.05). Grade Ⅲ gliomas had higher expression than gradeⅡsamples(P<0.05).Conclusion: Abnormal expression of MAPK and pMAPK indict the altered cell biological behavior and correlated to pathological grades of gliomas.
