摘要:
研究甲泼尼龙干预对缺血再灌注损伤(IRI)心肌组织中p53、caspase3mRNA表达的影响。方法:成年SD大鼠70只,随机分为3组。药物组(n=30)制备心肌IRI模型,然后右股静脉注入甲泼尼龙30mg·kg-1。IRI组(n=30)和假手术组(n=10)于右股静脉注入灭菌生理盐水0.75mg·kg-1。IRI组和药物组分别于缺血0.5h、缺血再灌注(IR)2、4、8、12和24h时各取5只大鼠处死,假手术组于缺血再灌注12h时处死动物,取出心脏,采用TUNEL法检测凋亡细胞,原位杂交法检测p53、caspase3mRNA。结果:IRI组与药物组p53mRNA和caspase3mRNA均于心肌缺血0.5h后即出现表达,p53mRNA在IR8h达高峰,而caspase3mRNA的峰值出现在IR12h,凋亡细胞计数在IR12h达高峰(P<0.05)。与IRI组同时相点比较,药物组p53、caspase3mRNA表达及凋亡细胞百分比均降低(P<0.05)。结论:p53和caspase3在缺血诱导的心肌细胞凋亡中发挥重要作用;甲泼尼龙可抑制细胞凋亡,对心肌IRI具有一定的保护作用。
Abstract:
To study the expression of p53 mRNA,caspase3 mRNA in myocardia tissue and the effects of methylprednisolone on them after myocardial ischemia reperfusion injury(MIRI).Methods:A total of 70 SD rats were randomly divided into 3 groups: control group(n=10), model group(n=30) and administration group(n=30).The activity and administration group of apoptosis cells and p53 mRNA,caspase3 mRNA in 3 groups were determined.Results: The peak value of apoptosis cells in model group and administration group were at IR 12 h, p53 mRNA and caspase3 mRNA increased along with the processs of IR,to the peak value at IR 12 h.The number of apoptosis cell,the expression of p53,caspase3 mRNA in administration group were lower than those in model group(P<0.05). Conclusion: Both p53 and caspase3 play an important role in cardiomyocytes apoptosis caused by ischemia,and methylprednisolone may inhibit cardiomyocytes apoptosis.
