摘要:
观察17β雌二醇(E2)对人子宫内膜腺癌Ishikawa细胞增殖、细胞周期及丝裂原活化蛋白激酶(MAPK)通路相关蛋白P21ras和pErk表达的影响,初步探讨雌激素通过MAPK通路促进子宫内膜癌形成的可能性。方法:运用MTT法检测不同浓度(0、10-10、10-8、10-6和10-4mol/L)E2作用不同时间(24、48、72h)后Ishikawa细胞的增殖情况;流式细胞技术检测不同浓度(0、10-10、10-8mol/L)E2作用24h后细胞周期的变化。免疫细胞化学检测10-6mol/L的E2作用于细胞不同时间(0、5、15、30min及1、2h)后P21ras和pErk蛋白的表达,不同浓度E2(0、10-10、10-8、10-6和10-4mol/L)作用于该细胞最佳时间后2种蛋白的表达情况。结果:随着E2浓度增加,Ishikawa细胞吸光度值上升并呈时间依赖性(P<0.05),细胞周期中G0/G1期比例下降、S期比例升高(P均<0.05)。10-6mol/L的E2作用于Ishikawa细胞30min时P21ras和pErk的活化最显著[(74.00±2.54)%,(68.00±4.81)%](P<0.05)。随着E2浓度增加P21ras和pErk表达均逐渐增加,呈剂量依赖性(P<0.05)。且P21ras和pErk蛋白的表达成正相关(P<0.05)。结论:雌激素可以促进子宫内膜癌Ishikawa细胞增殖和周期的进展,而且可以通过非转录效应迅速激活子宫内膜癌细胞中MAPK信号传导通路。
Abstract:
To observe the influence of 17βestradiol on proliferation, cell cycle progression, expressions of P21ras and pErk in endometrial carcinoma Ishikawa cells and to explore the possibility of endometrial carcinoma induced by estradiol.Methods:The effects of estradiol on proliferation, cell cycle phase of endometrial carcinoma Ishikawa cells were detected using MTT assay and fluorescenceactivated cell sorting technique.The activity of P21ras and pErk were examined by immunoprecipitation in Ishikawa cells after stimulation with 10-6 mol/L E2 for different times(0,5,15,30 min, and 1,2 h) to observe the optimal time and with varied doses of E2(0,10-10,10-8,10-6,10-4 mol/L)for the optimal time.Results:With increased concentrations of E2, the proliferation of Ishikawa cells increased in a timedependent manner(P<0.05). Cell cycle phase analysis revealed that percentage of Ishikawa cells at G0G1 phase (P<0.05) decreased and percentage of S phase cells increased significantly(P<0.05).The maximal activation of P21ras and pErk took place at 30 min in Ishikawa cells [(74.00±2.54)%,(68.00±4.81)%] after stimulation with 10-6 mol/L E2(P<0.05).With increased doses of E2,the expression of P21ras and pErk increased in a concentrationdependent manner(P<0.05). Between the expressions of P21ras and pErk, there was significant positive correlationship(P<0.05).Conclusion: 17βestradiol not only could promote the cell proliferation and the cell cycle progression of endometrial carcinoma Ishikawa cells,but also activate promptly MAPK signaling pathway by nonnuclear actions in endometrial carcinoma cells.
