摘要:
观察β榄香烯乳对人舌鳞状细胞癌Tca8113细胞体外放射敏感性的影响,并探讨其相关机制。方法:集落形成法观察β榄香烯乳对Tca8113细胞的放射增敏作用:对照组(0mg/L)、实验A、B组(40、60mg/L)细胞经β榄香烯乳作用24h后,分别进行0、2、4、6、8Gy的剂量照射1min,14d后进行集落计数。对照组、实验组β榄香烯乳作用Tca8113细胞24h后,流式细胞术分析作用前后细胞周期及凋亡变化,免疫细胞化学染色观察Bcl2、Bax蛋白表达变化。结果:2~8Gy照射后实验组(40、60mg/L)集落计数(个)分别为:(60.67±3.56)、(52.67±3.44);(50.00±2.45)、(49.39±6.15);(39.00±3.74)、(25.00±2.53);(31.00±2.37)、(13.00±2.00)。对照组(0mg/L)集落计数(个)为:(74.17±3.60)、(55.17±4.59)、(48.00±2.90)、(38.50±3.62)。3组比较差异均有统计学意义(P均<0.05)。实验组(40、60mg/L)凋亡率分别为(10.00±4.95)%、(18.50±6.57)%,对照组为(0.42±0.33)%,3组比较差异均有统计学意义(P均<0.05)。实验组(40、60mg/L)G2/M期细胞百分比分别为(18.30±6.45)%、(30.90±7.94)%,对照组为(4.40±2.61)%,3组比较差异均有统计学意义(P均<0.05)。β榄香烯乳作用Tca8113细胞后,凋亡抑制蛋白Bcl2的表达减弱,凋亡促进蛋白Bax的表达增强,3组比较差异均有统计学意义(P均<0.05)。结论:β榄香烯乳在低细胞毒性浓度下对Tca8113细胞有放射增敏作用;其机制可能是诱导凋亡和促使细胞G2/M期阻滞。
Abstract:
To detect the influences of βelemene on radiosensitivity of human squamous cell carcinoma of tongue Tca8113 cells cultured in vitro and disscuss the relative mechanism.Methods:Effect of radiosensitivity was observed with the methed of clone forming assay: control group(0 mg/L)and trial group(40,60 mg/L) Tca8113 cells were exposed to βelemene for 24 h and radiated by 0, 2, 4, 6, 8 Gy dose for 1 min,then were cultured in the common medium for 24 h and the number of cell clone was measured. After control group and trial group Tca8113 cells were exposed to βelemene for 24 h, change of cell cycle and apoptosis was analysed with flowcytometry(FCM).Results:Cell clone results of trial group were (60.67±3.56),(52.67±3.44);(50.00±2.45),(49.39±6.15);(39.00±3.74),(25.00±2.53);(31.00±2.37),(13.00±2.00).Control group(0 mg/L) were (74.17±3.60),(55.17±4.59),(48.00±2.90),(38.50±3.62). There was significant difference among three groups (P<0.05).Apoptosis rates of trial group were (10.00±4.95)%, (18.50±6.57)%, and control group was (0.42±0.33)%, there was significant difference among three groups (P<0.05).G2/M phase rates of trial group were (18.30±6.45)%, (30.90±7.94)%, and control group was (4.40±2.61)%,there was significant difference among three groups (P<0.05).After treatment with βelemene, Bcl2 expression of Tca8113 cells decreased and Bax expression increased among three groups(P<0.05).Conclusion: There may be effects of radiosensitivity of Tca8113 cells exposed to lowcytotoxic concentrations.The mechanisms may be apoptosis and arrest of G2/M phase.
