摘要:
研究天麻素对海人酸干预的神经干细胞分化的影响及其机制。方法:体外分离并培养新生Wistar大鼠神经干细胞,通过免疫组织化学法和免疫荧光法进行鉴定。将神经干细胞分为3组:第1组不加药为空白对照组,第2组加入不同浓度梯度的海人酸,第3组加入天麻素和不同浓度梯度海人酸,计算分化后神经元和胶质细胞比例。结果:培养的细胞具有自我更新和向神经元、少突胶质细胞和星形胶质细胞分化的潜能,是神经干细胞。海人酸可以有效诱导神经干细胞向胶质细胞分化,天麻素可在一定程度上阻断海人酸的作用,诱导神经干细胞向神经元分化。结论:天麻素可减少海人酸兴奋性的影响,诱导神经干细胞向神经元分化。
Abstract:
To study the influence of gastrodin on the differentiation of neural stem cells mediated by kanic acid.Methods:The neural stem cells from newborn Wistar rat were isolated and cultured in vitro, and then identified by immunofluorescence and immunohistochemistry method.The neural stem cells were allocated into 3 groups:the first group was normal control group,the second group was given kanic acid at different doses,and the third group,gastrodin and kanic acid at different doses.
