摘要:
比较淀粉样前体蛋白(APP)转基因小鼠与同遗传背景的正常鼠的神经干细胞在分化过程中细胞凋亡、Tau蛋白变化及Tau[pS396]磷酸化的情况。方法:剪取新生的APP转基因小鼠尾尖,常规提取DNA,用PCR法检测动物是否携带有APP基因;而后分别取APP+鼠和正常鼠海马部位脑组织,按常规进行神经干细胞(NSC)的培养,并将纯化的NSC进行诱导分化,根据细胞来源不同分为APP+细胞组和正常细胞组,用线粒体膜电位法检测APP+细胞组、正常细胞组的细胞凋亡情况,用免疫荧光化学和免疫蛋白印记法测定2组细胞总Tau蛋白水平及Tau[pS396]磷酸化的变化。结果:APP+组细胞凋亡率明显高于APP-细胞(P<0.05),APP+细胞中总Tau蛋白和Tau[pS396]的磷酸化水平明显高于APP-细胞组(P<0.05).
Abstract:
To explore cell apoptosis and the tau protein phosphorylated in the process of differentiation of neural stem cells from APP transgenic mice.Methods:Extracting of DNA from tails of newborn transgenic mouse was used to check for APP gene by PCR. Neural stem cells were separated from hippocampal of APP positive mice and APP negative mice mechanically and were cultured and induced to differentiate in vitro. The differentiated cells were divided into 2 groups: APP+ cell group and normal cell control group.
