摘要:
观察C2神经酰胺对牛晶状体上皮细胞(lens epithelial cells,LECs)凋亡的影响及半胱天冬酶3(Caspase3)活性的变化。方法:取第二代牛LECs,经不同浓度(25 μmol/L, 50 μmol/L,100 μmol/L)C2神经酰胺作用12 h,行TUNEL染色,荧光显微镜下计数凋亡细胞,计算凋亡率;Caspase3活性检测试剂盒检测牛LECs中Caspase3活性的变化。结果:随着C2神经酰胺浓度的增加,牛LECs凋亡率也随之升高(P<0.05),呈剂量依赖关系。随着C2神经酰胺浓度的增加,牛LEC中Caspase3活性的表达也随之增强(P<0.05)。结论:C2神经酰胺可以诱导牛LECs发生凋亡;在此过程中,存在Caspase3活性表达。
Abstract:
To study the effects of C2ceramide on the apoptosis of cattle lens epithelial cells(LECs), and observe the variation of Caspase3 activity.Methods:The second generation LECs were treated with different concentrations(25 μmol/L, 50 μmol/L,100 μmol/L) of C2ceramide for 12 h. After TUNEL staining,the rate of cell apoptosis was calculated; the variation of Caspase3 activity was determined by Caspase3 activity assay kit.
