摘要:
研究人乳头瘤病毒16型(HPV16)DNA阳性乳腺良、恶性病变组织中E6和E7蛋白的表达情况。方法:以经PCR筛选证实的HPV16 DNA阳性的56例乳癌和41例乳腺良性病变组织为研究对象,采用Western blotting方法检测HPV16 E6和E7蛋白。结果:56例HPV16 DNA阳性乳癌组织中,HPV16 E6、E7和E6/E7共表达阳性率分别为67.9%(38/56)、75.0%(42/56)和57.1%(32/56);41例HPV16 DNA阳性乳腺良性病变组织中,HPV16 E6、E7和E6/E7共表达阳性率分别为36.6%(15/41)、43.9%(18/41)和24.4%(10/41)。HPV16 DNA阳性乳癌组织中E6、E7和E6/E7共表达阳性率均高于乳腺良性病变组织(χ2分别为9.34,9.70,10.34,P均<0.01)。HPV16 DNA阳性乳癌组织中E6、E7和E6/E7共表达阳性率与患者年龄和肿瘤组织学类型无关(P均>0.05),但与淋巴结转移有关(χ2分别为3.88,15.86,9.34,P均<0.05)。结论:HPV16 E6和E7蛋白在乳癌发生发展中具有重要作用,并与乳癌淋巴结转移有关。
Abstract:
To investigate the expressions of human papillomavirus 16 type (HPV16) E6 and E7 proteins in HPV16 DNApositive breast benign and malignant lesions.Methods:A total of 56 HPV16 DNApositive breast carcinoma tissue and 41 breast benign lesions confirmed by PCR were used to detect HPV16 E6 and E7 proteins using Western blotting assay.Results: In 56 HPV16 DNApositive breast carcinoma tissue, the positive rates of HPV16 E6,E7, and E6/E7 coexpression were 67.9% (38/56), 75.0% (42/56), and 57.1%(32/56), respectively, all higher than those of 36.6% (15/41), 43.9% (18/41), and 24.4% (10/41), respectively, in 41 HPV16 DNApositive breast benign lesions(χ2 was 9.34,9.70,10.34,respectively,P<0.01). The HPV16 E6, E7, and E6/E7 coexpression were associated with lymph node metastasis of patients with breast carcinoma (χ2 was 3.88,15.86,9.34,respectively,P<0.05), but not with age or histological classification (P>0.05).Conclusion:HPV16 E6 and E7 proteins may play an important role in the carcinogenesis and progression, and were associated with lymph node metastasis of breast carcinoma.
