摘要:
观察人脐血间充质干细胞(HUCBMSCs)在体内诱导分化为神经细胞(神经元和神经胶质细胞)。方法:健康孕妇顺产脐血经分离、培养得到HUCBMSCs,移植于自由落体撞击法制作的大鼠脑创伤模型脑损伤区(移植组),另设假损伤、损伤、假移植组,每组12只大鼠。分别于移植后2周和4周,对植入脑内的HUCBMSCs应用免疫组织化学方法进行胶质纤维酸性蛋白(GFAP)和神经元特异性烯醇化酶(NSE)染色。结果:移植后2周和4周在大鼠脑移植区均发现GFAP、NSE阳性细胞,且2周时NSE阳性细胞数为(27.0±3.3)个/HP,高于4周时的(16.0±2.5)个/HP(t=5.31,P<0.05);4周时GFAP阳性细胞数为(23.0±3.2)个/HP,高于2周时的(12.0±2.1)个/HP(t=3.14,P<0.05)。其他3组均未见NSE、GFAP阳性细胞。结论:HUCBMSCs移植入脑创伤区后,可以向神经样神经元、神经胶质细胞分化。
