摘要:
探讨真核起始因子4E(eIF4E)反义寡核酸(ASODN)对人食管癌EC1细胞中eIF4E表达的影响。方法:将针对eIF4E不同基因位点的2条反义寡核苷酸(ASODN1、2)和1条无义寡核苷酸(NODN)转染EC1细胞(设5、10、15μmol/L3个转染浓度),分别培养24、48、72h,并设空脂质体转染作为空白对照,采用免疫细胞化学和原位杂交方法检测转染后细胞中eIF4E蛋白和mRNA表达的变化。结果:ASODN1、2转染EC1细胞培养24、48、72h后,各组中eIF4E蛋白和mRNA的表达均降低,2者分别与空白对照组相比,差异均有统计学意义(P<0.05);ASODN1、2组每个时间段内随浓度增加ASODN抑制效应增强,差异具有统计学意义(F=9.423、10.284、10.496、7.621、8.420和9.089,P<0.05),组间相同时间点、相同浓度的抑制效应比较差异无统计学意义(P>0.05);NODN组和空白对照组对eIF4E的表达均无抑制效应。结论:特异性的ASODN能有效抑制人食管癌EC1细胞中eIF4E的表达。
Abstract:
To investigate the effects of antisense oligodeoxynucleotides of eukaryotic initiation factor 4E(eIF4E)on the expression of eIF4E in human esophageal carcinoma EC1 cells.Methods:EC1 cells were transfected by two ASODNs and one NODN against different heparanase gene sites at different concentrations (5,10,15 μmol/L) for 24,48 and 72 h, respectively.Empty liposome group was used as control. The expression of eIF4E protein and mRNA was detected using immunohistochemistry and in situ hybridization.Results: Compared with Control group and NODN group,there was a significant decrease in the eIF4E protein and mRNA level in specific ASODN groups,and the inhibition effect was in a concentrationdependent manner.There were no inhibition effects observed in NODN and control groups.Conclusion:Specific ASODN can effectively inhibit the expression of eIF4E in EC1 cells.
