摘要:
探讨5氟尿嘧啶(5FU)和替尼泊苷(VM26)联合应用对胃癌BGC823细胞的体外抑制和诱导细胞凋亡的作用。方法:采用MTT法测定5FU单药(15.63、31.25、62.50、125.00、250.00mg/L),VM26单药(1.25、2.50、5.00、10.00、20.00mg/L)及联合用药(15.63/1.25、31.25/2.50、62.50/5.00、125.00/10.00、250.00/20.00mg/L)对BGC823细胞的体外生长抑制率,应用流式细胞仪测定15.63mg/L5FU、1.25mg/LVM26单药及其联合用药0、12、24、48h各时间点的细胞周期和凋亡率。结果:VM26和5FU单药及联合用药对胃癌BGC823细胞的生长抑制率随药物质量浓度的增加而增加。15.63mg/L5FU单药处理后12、24h时S期细胞增多,G2/M期细胞减少;1.25mg/LVM26单药处理细胞后G2/M期细胞随时间的延长逐渐增多;联合用药24h后S期细胞显著减少,G2/M期细胞显著增多;联合用药处理后12、24h细胞凋亡率比单药5FU和VM26处理相同时间点显著增加。结论:5FU和VM26单药均可抑制胃癌BGC823细胞的生长,低剂量联合用药抑制作用较单药应用增加。单药和联合用药均可诱导细胞凋亡,且细胞周期的分布不同。
Abstract:
To investigate the effects of 5FU and VM26 on the proliferation and apoptosis of gastric cancer cell line BGC823.Methods:MTT assay was used to examine the inhibition rate of cell growth when BGC823 cells were treated with various concentrations of 5FU(15.63,31.25,62.50,125.00,250.00 mg/L),VM26(1.25,2.50,5.00,10.00,20.00 mg/L) single and combination for 48 h; the distribution of cell cycle phases and the apoptosis rate were examined by flow cytometry.Results: The inhibition rate of cell growth was enhanced in a dosedepandent manner when BGC823 cells were treated with various concentrations 5FU,VM26 single or combined.When BGC823 cells were treated with 1.25 mg/L VM26,the cell cycle was arrested in the G2/M phase at 48 h.When BGC823 cells were treated with 15.63 mg/L 5FU,the cell cycle was arrested in the S phase at 12,24 h. When 1.25 mg/L VM26 was combined with 15.63 mg/L 5FU,the distribution of the cell cycle phase was different from that of single drug.The apoptosis rate in joint drug group at 12,24 h was higher than that in single drug group.Conclusion:5FU and VM26 single can inhibit the growth of BGC823.The inhibition rate of the lowdose joint group is higher than the single drug.5FU and VM26 single and combination both can induce apoptosis,but induce different cell cycle phase distribution.
