摘要:
探讨低剂量替尼泊苷(VM26)联合沙利度胺对人胃癌BGC823细胞裸鼠移植瘤生长、荷瘤小鼠骨髓造血功能的变化及新生血管形成的影响。方法:建立人胃癌BGC823细胞裸鼠移植瘤模型,建模成功后随机分为沙利度胺组、高剂量VM26组、低剂量VM26组、低剂量VM26联合沙利度胺组和对照组,每组各10只。经尾静脉注射不同剂量VM26、生理盐水,经口灌注沙利度胺。用药2周后测量裸鼠移植瘤瘤重,计算抑瘤率;观察裸鼠骨髓造血功能,检测移植瘤组织中血管内皮生长因子(VEGF)、CD44v6的表达以及微血管密度(MVD)的变化。结果:各组裸鼠移植瘤瘤重差异有统计学意义(F=24.570,P<0.05)。各组移植组织中VEGF、CD44v6表达及MVD的差异均有统计学意义(χ2/F=22.881、26.371、19.518,P均<0.05);各治疗组VEGF的表达和MVD均较对照组显著降低,低剂量VM26联合沙利度胺组CD44v6的表达较对照组显著降低。低剂量VM26联合沙利度胺组对骨髓造血功能的抑制较高剂量VM26组减轻。结论:低剂量VM26联合沙利度胺能抑制人胃癌BGC823裸鼠移植瘤的生长、新生血管的形成,且对骨髓的造血功能无明显影响。
Abstract:
To assess the effect of lowdose VM26 combined with thalidomide on growth and neoplastic angiogenesis of transplanted human gastric adenocarcinoma BGC823 tumor and the change of the bone marrow hematopoietic function in nude mice.Methods:A nude mouse treansplanted human gastric adenocarcinoma BGC823 tumor was established.The tumorbearing mice were treated with VM26 at different doses,thalidomide and lowdose VM26 combined with thalidomide.The bone marrow hematopoietic function in nude mice,the tumor weight,inhibition rates were evaluated after the mice were sacrificed.The expressions of VEGF,MVD and CD44v6 in tumor tissue were examined by immunohistochemistry staining.Results: Compared with control group, growth of tumor was significantly reduced in mice treated with VM26, thalidomide single and combined(F=24.570,P<0.05). The expressions of VEGF protein and MVD in tumor tissue decreased significantly in treatment groups(χ2/F=22.881,26.371,19.518,P<0.05). Compared with control group,MVD and the expression of VEGF in treatment groups decreased and the expression of CD44v6 in combined group decreased.Compared with the highdose teniposide,the lowdose VM26 combined with thalidomide decreased the restraint of bone marrow hematopoietic function.Conclusion:Lowdose VM26 combined with thalidomide can inhibit the growth and angiogenesis of transplanted tumor in nude mice.The combined group has decreased the injury of the bone marrow hematopoietic function in nude mice.
