摘要:
观察金莲花黄酮对K562、HeLa、Ec109、NCIH446肿瘤细胞增殖的影响。方法:①采用CCK8法检测0.793、1.586、3.172、6.344、9.516、12.688g/L金莲花黄酮对K562、HeLa、Ec109、NCIH446肿瘤细胞株增殖的影响。②采用流式细胞术观察0.793、1.586、3.172g/L金莲花黄酮对K562细胞周期的影响。结果:①金莲花黄酮在0.793~12.688g/L范围内对体外培养的K562、HeLa、Ec109、NCIH446肿瘤细胞生长均有明显的抑制作用,且抑制作用呈剂量依赖性(F=6.72,9.81,8.54,4.83,P均<0.05)。②金莲花黄酮可使K562G0/G1期细胞增多,S期细胞减少(F=12.56,20.86,P均<0.05)。结论:金莲花黄酮对体外培养的K562、HeLa、Ec109、NCIH446肿瘤细胞株的增殖均有很强的抑制作用;金莲花黄酮可使体外培养的K562细胞周期阻滞在G0/G1期,可能是其抑制K562肿瘤细胞增殖的机制。
Abstract:
To observe the effects of Trollius flavonoids on proliferation of K562,HeLa,Ec109,and NCIH446 tumor cells.Methods:By the method of CCK8,the effects of Trollius flavonoids with the concentration of 0.793,1.586,3.172,6.344,9.516,12.688 g/L on proliferation of K562,HeLa,Ec109,and NCIH446 tumor cells were detected. By flow cytometry,the effects of Trollius flavonoids with the concentration of 0.793,1.586 and 3.172 g/L on K562 cell cycle were observed.Results:Trollius flavonoids at 0.793~12.688 g/L had marked inhibitory effect on proliferation of 4 tumor cells in a dosedependent manner (F=6.72,9.81,8.54,4.83,P<0.05). Trollius flavonoids increased G0/G1 phase K562 tumor cells and reduced S phase cells(F=12.56,20.86,P<0.05).Conclusion:Trollius flavonoids have apparently inhibitory effects on the 4 tumor cells cultured in vitro. Cell cycle of K562 cell was blocked at G0/G1 phase,which may explain the inhibitory effects of Trollius flavonoids on proliferation of K562 cells.
