摘要:
观察苦瓜蛋白MAP30对大肠癌细胞株LoVo细胞生长及凋亡的影响。方法:采用MTT比色法分别观察20、40、60、80和100mg/LMAP30处理48h及40mg/LMAP30处理24、36、48和72h对LoVo细胞生长的影响,计算细胞生长抑制率。采用流式细胞仪检测40mg/LMAP30处理24h对LoVo细胞凋亡的影响。采用Northernblot和Westernblot分别检测40mg/LMAP30作用于LoVo细胞0~120h,细胞bcl2和bax基因和蛋白的表达。结果:MAP30可呈剂量、时间依赖性抑制LoVo细胞的生长(F=128.3,107.5,P<0.05),促进LoVo细胞凋亡(F=92.7,P<0.05)。MAP30还可上调Bax基因与蛋白表达,下调Bcl2基因与蛋白的表达。结论:MAP30可影响LoVo细胞Bcl2和Bax的表达,从而诱导细胞凋亡,抑制细胞生长。
Abstract:
To study the effects of MAP30 on cell growth and apoptosis of LoVo cells and to elucidate its molecular mechanism.Methods:LoVo cells were allocated into 6 groups and treated with 0,20,40,60,80,and 100 mg/L MAP30 for 24 h.Another LoVo cells were allocated into 4 groups and treated with 40 mg/L MAP30 for 24,36,48,72 h. MTT assay was used to detect the inhibitory rate of cell growth.Flow cytometry was used to detect the apoptosis of LoVo cells treated with 40 mg/L MAP30 for 24 h.Northern blot and Western blot were used to detect the mRNA and protein expression of Bcl2 and Bax.Results: MAP30 inhibited the cell growth of LoVo in a dose and timedependent manner (F=128.3 and 107.5,P<0.05),and induced the apoptosis of LoVo cells(F=92.7,P<0.05).MAP30 could upregulate the mRNA and protein expression of Bax,and downregulate the mRNA and protein expression of Bcl2.Conclusion:MAP30 can inhibit cell growth and induce apoptosis of LoVo cells, which may be realized by regulating the expression of Bcl2 and Bax.
