摘要:
观察姜黄素对人肝星状细胞LX2形态学的影响。方法:通过HE染色及丫啶橙荧光染色观察LX2细胞经 0(对照)、10、20、30、40及50 μmol/L的姜黄素作用12 h后的形态学变化。结果:①倒置显微镜下可见对照组细胞呈成纤维细胞样表型,而姜黄素组细胞数变少,细胞变小,形态向椭圆形或圆形改变,胞质浓缩,且随姜黄素浓度的增加变化更加明显。②HE染色光镜下观察显示,姜黄素组凋亡细胞增加,表现为胞体缩小、变圆,胞核固缩、碎裂,染色体被染成蓝黑色。③丫啶橙染色激光共聚焦显微镜下可见明显的凋亡细胞的多种形态改变,凋亡小体清晰可见。结论:经姜黄素作用人肝星状细胞可出现明显的增殖受抑、细胞凋亡的形态学改变。
Abstract:
To investigate the effects of curcumin on the morphology of human hepatic stellate cell(HSC) line LX2.Methods:The morphological changes of human HSC treated by different concentrations (0,10,20,30,40,and 50 μmol/L) of curcumin for 12 h were observed by inverted microscopy,HE staining, and Acridine orange staining.Results:The inverted microscopy revealed that the human HSC population lessened, cells grew smaller, and shape of cells became round or oval gradually,and these changes were related with the concentration of curcumin.HE staining revealed that the apoptotic cells increased after curcumin treatment. Smaller and rounder cell bodies,fragmentation and pycnosis of black nuclei were found in the apoptotic cells.Acridine orange staining indicated there were conspicuous morphologic changes of the apoptotic cells after curcumin treatment. Apoptotic bodies could be found easily.Conclusion: After being treated with curcumin, typic morphologic changes of proliferative inhibition and apoptosis of human HSC could be identified.
