摘要:
检测生理 pH值下偏端霉素衍生物与DNA 间的相互作用。方法:单独的配体和DNA在300~380 nm区域都没有信号,向DNA液中滴加偏端霉素衍生物,在圆二色谱(CD谱)上300~370 nm区域会有信号出现。利用这种方法观察核酸构象的变化。结果:在偏端霉素衍生物与DNA的混合液的CD谱上观察到在300~370 nm区域有特征峰。结论:偏端霉素衍生物对DNA的碱基有识别功能。
Abstract:
To detect the biological activity of new synthetic distamycin derivatives, the present work gives the results of studies on the interaction between distamycin derivatives and DNA under physiological pH.Methods:Neither the free DNA nor the free ligands exhibited circular dichroism(CD) signals in the 300~380 nm wavelength region. With incremental titration of distamycin derivatives, in the CD spectra of mixed solutions a specific absorption peak will appeare in the 300~380 nm wavelength region. By this method to study the structure change of DNA.Results: In the CD spectra of the mixing solution of DNA with distamycin derivatives a specific peak appeared in the region of 300~370 nm wavelength.Conclusion:The distamycin derivatives may have ability to recognize the base of DNA.
