摘要:
制备氟甲砜霉素(FF)的最佳人工抗原并鉴定。方法:采用琥珀酸酐将FF的羟基活化后制备出半抗原氟甲砜霉素半琥珀酸(FFHS),并经质谱(MS)鉴定合成成功。采用碳二亚胺法将FFHS与牛血清白蛋白(BSA)偶联,采用混合酸酐法将FFHS与卵清白蛋白(OVA)偶联,采用羰基二咪唑法将FF直接与 BSA偶联,制备了3种人工抗原(FFHSBSA、FFHSOVA和FFBSA)。采用紫外吸收光谱分析和动物免疫实验(3种偶联物选择前2种分别作为免疫原与包被原进行抗血清制备及 ELISA实验)鉴定3种人工抗原。结果与结论:紫外吸收光谱分析初步证实3种人工抗原合成成功。在动物免疫实验中FFHSBSA 与 FFHSOVA得到了效价较高的特异性抗体,IC50分别为150、300 μg/L。FFBSA免疫原免疫效果不理想。
Abstract:
To prepare and identify florfenicol artificial antigen.Methods: Florfenicol was actived by succinic anhydride and converted into florfenicol succinate as the hapten.The result of MS indicated that the hapten was synthesized successfully. Florfenicol succinate was linked to carrier protein bovine serum albumin(BSA) and ovalbumin(OVA),by carbodiimide method, mixed anhydride method,respectively,and florfenicol was linked to carrier protein BSA as artificial antigen by N,N′carbonyldiimidazole(CDI) method.Antigens FFHSBSA, FFHSOVA, and FFBSA were prepared and confirmed by ultraviolet spectrum and animal experiment. Results and Conclusion: The 3 antigens were confirmed to be synthesized successfully by ultraviolet spectrum. The antiserum produced by FFHSBSA and FFHSOVA showed special sensitivity to the florfenicol,and IC50 was 150,300 μg/L, respectively.
