摘要:
探讨利用超顺磁性氧化铁颗粒(SHU555A)体外标记大鼠骨髓间充质干细胞(BMSCs)对细胞活力的影响。方法:分别使用不同铁浓度(35、70、140和210 mg/L)的SHU555A标记大鼠BMSCs,分别于标记后1 d及1、2、3周行普鲁士蓝染色观察铁纳米颗粒进入细胞情况,台盼蓝排除试验检测细胞活力。结果:普鲁士蓝染色显示标记的大鼠BMSCs胞质内存在细小蓝色铁颗粒,标记率100%;台盼蓝染色显示标记时间对BMSCs活力无影响,铁浓度为35 mg/L时对标记的细胞活力无影响,大于该浓度则细胞活力下降。结论:35 mg/L的铁纳米颗粒可用于大鼠BMSCs标记。
Abstract:
To investigate the effects of super paramagnetic iron oxide particles (SHU555A) on the survival of rat bone marrow stem cells(BMSCs).Methods:BMSCs were labeled with different concentration (35,70,140 and 210 mg/L) of iron for 1 day, 1, 2, and 3 weeks,particles within BMSCs and viability of the labeled cells were evaluated by Prussian blue stain and trypan blue exclusion test.Results: BMSCs could be effectively labeled with a labeling efficiency of 100%.Prussian blue stain results showed that small blue iron particles exist in BMSCs, as well as trypan blue test showed that 35 mg/L concentration of iron particles had no effect on viability of the labeled cells, whereas concentration higher than 35 mg/L could decline the viability of BMSCs. Compared with nonlabeled cells, SHU555A labeled BMSCs did not have significant difference 1 day, 1, 2, and 3 weeks later.Conclusion:Iron partieles of 35 mg/L could be used to label BMSCs efficiency.
