摘要:
探讨内皮祖细胞(EPCs)自体移植联合阿托伐他汀治疗对急性心肌梗死(AMI)大鼠心功能的影响。方法:30只SD大鼠随机分为心肌梗死对照组(组Ⅰ)、单纯EPCs移植组(组Ⅱ)和阿托伐他汀联合EPCs移植组(组Ⅲ),每组10只。大鼠无菌心脏采血,应用密度梯度离心法分离单个核细胞,体外培养,获得较纯的EPCs。3组大鼠均结扎左冠状动脉前降支,制造大鼠AMI模型。组Ⅱ、组Ⅲ在模型制备后,于梗死周边区注射5溴2’脱氧尿嘧啶核苷(BrdU)标记的EPCs,组Ⅰ注射等量培养液。24 h后,组Ⅲ以阿托伐他汀50 mg/(kg·d)灌胃。8周后观察大鼠心功能及血流动力学指标的改变,并以免疫组化方法检测心肌组织内EPCs及修复情况。结果:8周后,3组左室短轴缩短率、射血分数、左室收缩压、左室舒张末压及压力变化速率最大值比较,差异均有统计学意义(F分别为95.7、296.5、844.9、67.6、127.0,P均<0.001)。组Ⅲ大鼠心功能及血流动力学指标较组Ⅱ和组Ⅰ改善(P均<0.05),缺血区域的移植细胞部分分化为血管内皮细胞。组Ⅰ大鼠心肌组织结构紊乱,心肌细胞被瘢痕组织替代;组Ⅱ和组Ⅲ心肌结构排列明显有序。结论:EPCs移植联合阿托伐他汀治疗AMI效果优于单纯EPCs移植。
Abstract:
To study the effects of autologous endothelial progenitor cell(EPCs) transplantation plus atorvastatin treatment on heart function of acute myocardial infarction(AMI) rats.Methods:A total of 30 SD rats were allocated into three groups randomly, the model group (groupⅠ),EPCs transplantation group (groupⅡ),and EPCs transplantation plus atorvastatin group(groupⅢ),with 10 rats in each group. Mononuclearcells collected from the heart blood of rats were isolated by density gradient centrifugation. The autologous EPCs were purified by culture in vitro, and labeled by BrdU. AMI was induced by ligating the left anterior descending coronary artery. Rats of groupⅡand groupⅢ received EPCs transplantation in the infarct perimeter areas. Rats of groupⅠreceived equal dose of culture medium. After 24 hours,rats of groupⅢ were lavaged with atorvastatin at dose of 50 mg/(kg·d). After 8 weeks, heart function and dynamic index were detected, EPCs in myocardium were examined by immunohistochemical staining,and the pathological changes of myocardium were observed.Results: After 8 weeks,there were significant differences in LVFS,LVEF,pLVS,pLVED and (dp/dt)max among the three groups (F=95.7,296.5,844.9,67.6,and 127.0,respectively,P<0.001).Heart function and dynamic index in groupⅢ were significantly improved(P<0.05), some of transplanted EPCs differentiated into vascular endothelial.Conclusion:The efficacy of EPCs transplantation plus atorvastatin for AMI is priors to EPCs transplantation lonely.
