摘要:
观察α硫辛酸对早期糖尿病性视网膜病变大鼠视网膜组织血管内皮生长因子(VEGF)和蛋白激酶C(PKC)表达的影响,探讨α硫辛酸防治早期糖尿病视网膜病变的作用机制。方法:雄性Wistar大鼠57只,其中42只采用链脲佐菌素一次性腹腔注射制作糖尿病模型,造模成功后随机分为α硫辛酸治疗组(19只)和糖尿病组(18只),另外15只大鼠为正常对照组。α硫辛酸治疗组隔 d给予α硫辛酸腹腔注射,糖尿病组及正常对照组给予等量生理盐水,连续12周,记录给药4、8及12周各组空腹血糖。12周后处死实验大鼠,取眼球,制作视网膜组织切片,采用免疫组织化学染色检测VEGF和PKC蛋白的表达。结果:①给药4、8及12周,3组大鼠空腹血糖水平相比,差异有统计学意义(F时间=3.21,P=0.02),其中α硫辛酸治疗组和糖尿病组空腹血糖水平均高于正常对照组(F组间=615.31,P<0.001),但2组间空腹血糖水平差异无统计学意义(P>0.05)。②给药12周后3组大鼠视网膜VEGF和PKC蛋白表达水平差异有统计学意义(F=74.02和169.69,P均<0.01),其中糖尿病组与α硫辛酸治疗组大鼠均较正常对照组VEGF和PKC蛋白表达水平上调(P均<0.05),但α硫辛酸治疗组视网膜组织VEGF和PKC蛋白表达水平较糖尿病组降低(P均<0.05)。结论:α硫辛酸可下调早期糖尿病大鼠视网膜组织中PKC和 VEGF蛋白的表达,干预早期糖尿病视网膜病变的发生、发展。
Abstract:
To investigate the roles and mechanisms of αlipoic acid in preventing and curing earlier period diabetic retinopathy.Methods:Out of 57 male healthy Wistar rats,42 rats were chosen randomly and injected the streptozotocin intraperitoneally once to set up diabetic model. After the diabetic model were established and randomly allocated into 2 groups: the diabetes group and the αlipoic acid treatment group. At the same time, 15 rats were injected citrate buffer solution intraperitoneally as the normal control group. αlipoic acid was given to the αlipoic acid treatment group by intragastric administration every other day;normal saline was given by intragastric administration to the diabetes group and the normal control group. All rats were regularly measured bloodfasting glucose at the end of the 4th week, 8th week and 12th week. After 12 weeks,rats were sacrificed,and retinal tissue was obtained for immunohistochemical staining to detect the expression of VEGF and PKC.Results:The bloodfasting,glucose of 3 groups at 3 time points had significant difference(Ftime=3.21,P=0.02),and the bloodfasting glucose of the αlipoic acid treatment group and the diabetic group higher than that of the normal control group(Fgroup=615.31,P<0.001),but there was no significant difference between the αlipoic acid treatment group and diabetic group (P>0.05).The expressions of VEGF and PKC of 3 groups at the end of the 12th week had significant difference(F=74.02 and 169.69,P<0.05). Compared with those of the normal control group,the expression of VEGF and PKC was higher in the αlipoic acid treatment group and the diabetic group(P<0.05),but that of the αlipoic acid treatment group was lower than that of the diabetic group(P<0.05).Conclusion: αlipoic acid could regulate down the expression of PKC and VEGF,which may interfer in the development of earlier period diabetic retinopathy.
