摘要:
观察葛根素(Pue)对牛主动脉内皮细胞(BAECs)增殖的影响。方法:取BAECs细胞,分为空白对照组(C组):常规培养24 h;溶媒组(S组):加PBS后培养24 h;葛根素(Pue)低、中及高剂量组:分别加入终质量浓度为0.5 g/L(P1组)、1.0 g/L (P2组)及1.5 g/L(P3组)的Pue,再培养24 h。另取细胞,同上分组,均于加药前2 h加入L单甲基精氨酸(LNMMA),加药后再培养24 h。采用MTT法检测细胞增殖情况,流式细胞仪检测细胞周期,Western Blot检测细胞内皮型一氧化氮合酶(eNOS)的表达。结果:Pue可促进BAECs的增殖,升高S+G2/M期的细胞比例,促进BAECs eNOS蛋白的表达(F=277.27,570.65,1 051.99,P<0.001),而LNMMA可抑制上述效应。结论:Pue可促进BAECs进入分裂期从而促进增殖,其机制可能与增加eNOS的表达从而上调NO水平有关。
Abstract:
To observe the effects of puerarin(Pue) on the proliferation of bovine artery endothelial cells (BAECs).Methods:Ⅰ:BAECs were cultured and allocated into group C,group S and low,middle and highdose Pue groups (group P1,P2, and P3).Group C was not treated;group S and group P1,P2 and P3 were given PBS, low,middle and highdose Pue and then cultured for 24 h. Ⅱ:BAECs were allocated into 5 groups as above but added LNMMA 2 h before administration.MTT and flow cytometry were used to detect cell proliferation and cell cycle, respectively.Western Blot was used to detect the expression of eNOS.Results:Pue could inhibit cell proliferation and increase percentage of S+G2/M cells and the expression of eNOS in a dose dependent manner(F=277.27,570.65,1 051.99,P<0.001).LNMMA could inhibit the effects of Pue(P<0.05). Conclusion:Pue can prompt the proliferation of BAECs,which may be involved in its increasing the expression of eNOS.
