摘要:
探讨氯化汞(HgCl2)对体外培养的小鼠睾丸间质瘤细胞mLTC1活性及孕酮合成的影响。方法:分别以10-4、10-5 、10-6、10-7、10-8、10-9 和10-10 mol/L HgCl2对mLTC1细胞进行染毒,24 h 后噻唑蓝(MTT)法观察mLTC1细胞活性;分别以10 kU/L人绒毛膜促性腺激素(HCG)混以0、10-8、10-7、10-6和10-5 mol/L的HgCl2对mLTC1细胞刺激2 h,放射免疫法测定mLTC1细胞孕酮的分泌量。结果:HgCl2在10-6 ~ 10-10 mol/L剂量范围内对mLTC1细胞活性无影响,10-5 mol/L HgCl2刺激细胞增殖,10-4 mol/L HgCl2 抑制细胞增殖(F=5.268,P<0.001)。随HgCl2剂量由0增至10-6 mol/L,mLTC1细胞孕酮分泌量逐渐下降(F=4.76,P<0.001)。结论:HgCl2可以直接影响mLTC1细胞活性及其孕酮的分泌。
Abstract:
To investigate the effects of mercury chloride(HgCl2 ) on the cell growth and progesterone biosynthesis in MA10 mouse Leydig tumor(mLTC1) cells.Methods:mLTC1 cells were exposed to 0, 10-10, 10-9, 10-8, 10-7 , 10-6 , 10-5 and 10-4 mol/L HgCl2 for 24 h, respectively, MTT was used to observe cells activity. mLTC1 cells induced by 10 kU/L HCG were exposed to 0, 10-8, 10-7 , 10-6 and 10-5 mol/L HgCl2 for 2 h, respectively, progesterone level in supernate were measured by RIA.Results: HgCl2 had no significant effects on the activity of mLTC1 cells at the concentrations of 10-10~10-6mol/L, but it could promote cells growth at the dose of 10-5 mol/L and significantly inhibit cells growth at the dose of 10-4 mol/L(F=5.268,P<0.001). Progesterone level in supernatethe decreased with HgCl2 concentration changing from 0 to 10-6 mol/L(F=4.76,P<0.001).Conclusion: HgCl2 could directly affect cell growth and the progesterone synthesis of mLTC1 cells.
