摘要:
探讨食管鳞癌组织和其细胞系的肿瘤干细胞(CSC)标志。方法:应用免疫酶或免疫荧光技术检测20例食管鳞癌组织和细胞(Eca109和EC9706)中CD133、CD44以及多药耐药基因1(MDR1)的表达和定位。结果:食管鳞癌细胞中CD133和MDR1一致强阳性的细胞占30%~43%,CD133和CD44一致强阳性的细胞约占40%。CD133强阳性小细胞占癌细胞的8%,癌组织中约为4%。CD133强阳性小细胞位于鳞癌上皮的基底层、癌珠周缘或癌灶内;CD44强阳性细胞约占40%,其中包含2个亚群:少数较小CD44阳性的细胞亚群定位于鳞癌上皮的基底层、癌珠周缘或癌灶内;较大CD44强阳性细胞的亚群主要定位于鳞癌上皮棘细胞层及癌灶中。结论:CD133强阳性小细胞和CD44强阳性细胞可能是CSC的标志。
Abstract:
To explore cancer stem cell (CSC) markers in human esophageal squamous cell carcinoma(ESCC).Methods:Expressions of CD133, multidrug resistance 1(MDR1)and CD44 were examined in 20 surgically resected ESCC tissues and Eca109 and EC9706 cells by immunoenzyme or immunofluorescence technique.Results: The percentage of CD133 and MDR1 positive cells wars 30%~43%,and the positive cells with CD133 and CD44 accounted for about 40% in Eca109 and EC9706 cells.The small cells (CD133+) were located in the epithelial basal layer, cancer focus and at cancer pearl periphery. In ESCC roughly 40% CD44+ cells included small cell subpopulation located in the epithelial basal layer, cancer focus and at cancer pearl periphery and larger cell subpopulation mainly located in the epithelial prickle layer and scattered in the cancer focus.Conclusion: In ESCC cells, CD133 and CD44 may be the marker of CSC.
