摘要:
观察苦菜总黄酮对大鼠心肌缺血再灌注损伤的保护作用。方法:30只SD大鼠随机分成5组,假手术组、模型组和苦菜总黄酮高(100mg/kg)、中(50mg/kg)、低(25mg/kg)剂量组,每组6只。结扎大鼠左冠状动脉前降支制备心肌缺血再灌注损伤模型。苦菜总酮各组实验前灌胃相应剂量的药物,模型组和假手术组灌胃质量分数0.5%羧甲基纤维素钠。显微镜镜下观察心肌坏死程度;比色法检测大鼠血清中乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)、丙二醛(MDA)和髓过氧化物酶(MPO)水平。结果:光镜下假手术组心肌纤维排列整齐;模型组可见片状心肌细胞变性坏死,肌纤维肿胀断裂,间质内大量炎性细胞浸润和出血;苦菜总黄酮组心肌病变较模型组减轻。5组间血清LDH、SOD、MDA和MPO含量比较,差异有统计学意义(F=45.633、38.623、4.840和22.658,P均<0.05);模型组血清LDH、MDA和MPO水平高于假手术组,SOD水平低于假手术组。苦菜总黄酮组血清LDH、MDA和MPO水平均低于模型组,血清SOD水平高于模型组。苦菜总黄酮高、中、低剂量组血清LDH、SOD、MOA和MPO含量两两比较,差异无统计学意义(P>0.05)。结论:苦菜总黄酮对心肌缺血再灌注损伤有保护作用。
Abstract:
To investigate the protective effects of sonchus oleracens flavonoids on myocardial ischemiareperfusion injury in rats.Methods:A total of 30 SD rats were randomly allocated into five groups(6 rats per group):sham operation group group,model group,low,medium,high dose sonchus oleracens flavonoids groups.The myocardial ischemiareperfusion injury model in rats was established by ligating left anterior descending coronary artery.The rats of sonchus oleracens flavonoids groups were given the medicine before experiment,while the control and model groups were taken carboxymethyl cellulose sodium.The contents of lactate dehydrogenase(LDH),superoxide dismutase(SOD),malondialdehyde(MDA),and myeloperoxidase(MPO) in serum were detected.Results: The normal control group myocardial fibers were aligned under microscope.There were lots of degeneration necrosis of myocardial cells and muscle swelling fracture, a lot of infiltrating Inflammatory cells,bleeding in myocardial cells in the model group.In sonchus oleracens flavonoids groups myocardial lesions were milder compared with the model group.Levels of serum LDH, SOD, MDA, and MPO among the groups were different,and there were obvious differences(F=45.633,38.623,4.840 and 22.658,P<0.05). Levels of serum LDH, MDA, and MPO in the model group were higher than the normal control group,those of the sonchus oleracens flavonoids groups were lower than the model group.The serum SOD levels in the model group were lower than that of the sonchus oleracens flavonoids groups.Levels of serum LDH, SOD, MDA, and MPO among the sonchus oleracens flavonoids high,medium,low dose groups had no statistical significance(P>0.05).Conclusion:Sonchus oleracens flavonoids exerts protective effects on myocardial ischemiareperfusion injury.
