摘要:
观察硫辛酸(LA)对肾缺血/再灌注(RIR)后心肌损伤的影响。方法:30只大鼠随机分为对照组、缺血/再灌注(IR)组和IR+LA组,IR组复制大鼠RIR模型,IR+LA组再灌前28d灌胃给予LA250mg/(kg·d)。观察3组大鼠心肌组织丙二醛(MDA)、髓过氧化物酶(MPO)、黄嘌呤氧化酶(XO)、超氧化物歧化酶(SOD)和血浆肌酸激酶(CK)、心肌型肌酸激酶同工酶(CKMB)及一氧化氮(NO)的变化;测定平均动脉压(pMA)、左室收缩压(pLVS)、收缩期左室内压上升最大速率(dp/dtmax)、舒张期左室内压下降最大速率(-dp/dtmax);免疫组织化学法测定心肌组织中Caspase3蛋白表达。结果:3组间MDA、MPO、XO、SOD、CK、CKMB、NO、Caspase3阳性细胞率、pMA、pLVS、dp/dtmax和-dp/dtmax比较,差异均有统计学意义(F分别为38.540、48.320、25.270、28.960、38.750、42.110、36.350、49.410、28.810、37.170、23.230和33.650,P均<0.05)。结论:LA可能通过抗炎、抗氧化、升高NO含量及抑制凋亡等途径对RIR后的心肌发挥保护作用。
Abstract:
To investigate the effects of lipoic acid (LA) on myocardium in rats suffered from renal ischemia reperfusion.Methods:A total of 30 rats were randomly divided into 3 groups:control group,ischemia reperfusion(IR) group,and IR+LA group (n=10).Models of renal IR were made.Contents of malondialdehyde(MDA), myeloperoxidase (MPO), xanthine oxidase (XO) and superoxide dismutase (SOD) in myocardia were measured. Levels of creatine kinase (CK) , creatine kinase MB isoenzyme (CKMB) and nitric oxide (NO) in plasma were determined. The mean arterial pressure (pMA), left ventricular systolic pressure (pLVS), maximal rise rate of left ventricular pressure (dp/dtmax),and the maximal fall rate of left ventricular pressure (-dp/dtmax) were monitored. The change of myocardial morphology and expression of Caspase3 were observed by light microscope and immunohistochemistry method.Results:Results indicated that MDA,MPO,XO,SOD,CK,CKMB,NO,percentage of Caspase3 positive cells,pMA,pLVS,dp/dtmax, and -dp/dtmax had difference among different groups(F was 38.540,48.320,25.270,28.960,38.750,42.110,36.350,49.410,28.810,37.170,23.230,and 33.650,P<0.05).Conclusion: By the way of antiinflammatory, antioxidation, increasing the content of NO,and inhibiting apoptosis, LA possibly protects the myocardia suffered from renal IR.
