摘要:
观察酸性肽对体外培养的大鼠星形胶质细胞增殖的影响。方法:SD大鼠来源的星形胶质细胞经传代纯化后分为5组培养:无血清组、体积分数20%的胎牛血清对照组和低、中及高剂量酸性肽组(分别给予37.5、75.0及150.0 mg/L酸性肽),作用24、48及72 h后,收集星形胶质细胞及上清液,计算细胞存活率,并采用日立7170全自动生化分析仪测定上清液中乳酸脱氢酶(LDH)的含量。另取细胞,同上分组后培养72 h,采用MTT法观察细胞增殖情况,计算细胞增殖率。结果:培养24、48和72 h后,5组细胞存活率及上清液中LDH含量差异均有统计学意义(细胞存活率F=149.381、58.817和119.135,LDH含量F=1 247.644、2 381.877和1 102.340,P均<0.001);5组不同时间点间细胞存活率及上清液中LDH含量差异均有统计学意义(细胞存活率F=62.777、50.993、10.555、38.874和21.742,LDH含量F=775.762、222.799、184.471、59.806和135.954,P均<0.001)。培养72 h后,5组细胞增殖率相比,差异有统计学意义(F=137.416,P<0.001)。与相同时间点的无血清组和胎牛血清对照组相比,酸性肽各组细胞存活率和增殖率均升高,上清液中LDH含量均降低(P均<0.05)。结论:酸性肽对体外培养的大鼠星形胶质细胞的增殖具有一定的营养和保护作用。
Abstract:
To research the influence of acidic neuropeptide on the proliferation of rat astrocytes in vitro.Methods:The purified astrocytes were allocated into 5 groups: serumfree group, 20% serum group and 3 different concentration of acidic neuropeptide groups(37.5, 75.0 and 150.0 mg/L). After being cultured for 24,48,and 72 h,the cells and supernatant were collected to detect the survival rate and LDH in supernatant by trypan blue dying and Rili 7170 fully automatic biochemistry analyzer,respectively. Astrocytes were allocated as above and cultured for 72 h, and the proliferation was observed by MTT assay.
