摘要:
目的:观察甲磺酸伊马替尼(IM)对游离二氧化硅(SiO2)粉尘刺激所致大鼠肺成纤维细胞(LF)激活过程的影响。方法:以游离SiO2粉尘工作液对大鼠肺泡巨噬细胞(AM)染毒24 h,离心收集其培养上清。阳性对照组用该培养上清刺激大鼠LF,3个实验组在培养上清中分别加入终浓度为0.2、1.0和5.0 μmol/L的IM后处理大鼠LF,阴性对照用未经SiO2刺激的AM上清培养LF;24 h后,收集各组培养上清,使用MTT法检测LF细胞存活情况,用RTPCR方法检测LF α平滑肌肌动蛋白(αSMA)、Ⅰ型胶原(COLⅠ)及Ⅲ型胶原(COLⅢ)mRNA的表达水平,用ELISA法测定培养上清中的COLⅠ和COLⅢ蛋白,用BradFord法测定培养上清中的总蛋白。结果:5组存活细胞比例、αSMA mRNA、COLⅠmRNA及蛋白、COLⅢ mRNA及蛋白和总蛋白表达水平差异均有统计学意义(F=3.571、3.904、18.686、6.582、28.983、8.555和9.918,P均<0.05)。与阴性对照组比较,阳性对照组上述指标均增高(P均<0.05)。与阳性对照组比较,各实验组上述指标均降低,且随IM浓度的升高而降低(P均<0.05)。结论:IM可抑制游离SiO2粉尘诱导大鼠LF的激活。
