摘要:
 目的:研究双点法注射6羟基多巴胺(6OHDA)制备的帕金森病(PD)大鼠相关分子的改变。方法:采用脑立体定位仪6OHDA微量注射建立大鼠PD模型,2周后注射阿朴吗啡(APO),观察其行为学改变,并利用高效液相电化学法测定正常大鼠(n=10)及PD大鼠(n=23)黑质组织中多巴胺(DA)及其代谢物3,4二羟基苯乙酸(DOPAC)的含量;分光光度法测定超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量;RTPCR法测定bcl2和bax mRAN的表达。结果:与正常大鼠比较,PD大鼠黑质组织中DA及其代谢物DOPAC的含量降低(t=17.069和17.087,P均<0.001),SOD 活性降低(t=6.314,P<0.001), MDA含量升高(t=11.060,P<0.001),bcl2 mRNA表达降低(t=9.333,P<0.001),而bax mRNA表达则升高(t=6.504,P<0.001)。结论:PD的发生与氧自由基损伤导致的神经元退变有关。
