摘要:
目的:建立分泌高水平白细胞介素6(IL6)的耐紫杉醇卵巢癌细胞株的小鼠移植瘤模型,评估抗IL6单克隆抗体Siltuximab(CNTO328)对异种移植肿瘤生长的影响。方法:选用3~4周龄的雌性裸小鼠36只,第1天用5×106 mL-1 SKOV3TR细胞和基质胶的混悬液作鼠皮下注射,2周后分成6组,分别给予腹腔注入生理盐水、非特异性抗体F105(20 mg/kg)、Siltuximab(20 mg/kg)、紫杉醇(20 mg/kg)、紫杉醇(20 mg/kg)和Siltuximab(20 mg/kg)混合液、紫杉醇(20 mg/kg)和非特异性抗体F105(20 mg/kg)混合液。每周治疗2次,持续5周。每天检查小鼠健康状况和肿瘤生长情况。结果:各组肿瘤体积比较,差异无统计学意义(F组间=2.820,P=0.310;F时间=3.020,P=0.230;F交互=2.950,P=0.280)。结论:Siltuximab对裸鼠体内卵巢癌耐药细胞SKOV3TR移植瘤的生长无明显影响。
Abstract:
Aim:To explore potential therapeutic strategies for interrupting the IL6 signaling through this pathway,xenograft model of chemoresistance ovarian cancer cell lines (secrete high level of IL6) was established,and evaluate Siltuximab effect on xenograft tumor growth.Methods:A total of 36 female nude mice at approximately 3 to 4 weeks of age were selected,5×106 mL-1 SKOV3TR cells were injected subcutaneously with matrigel on the first day.Two weeks after injection, the mice were randomly divided into 6 groups.Group A was received intraperitoneal injection with normal saline,Group B with nonspecific antibody F105 (20 mg/kg),and Group C with Siltuximab (20 mg/kg),Group D with paclitaxel (20 mg/kg),Group E with paclitaxel (20 mg/kg) and Siltuximab (20 mg/kg),and Group F received paclitaxel (20 mg/kg) and F105 (20 mg/kg). All six groups were treated twice a week for five weeks. The health of the mice and evidence of tumor growth were examined daily.Results:The xenograft tumor volume of the six groups had no significant difference (Fgroup=2.820,P=0.310;Ftime=3.020,P=0.230;Finteraction=2.950,P=0.280).Conclusion:Siltuximab does not have effect on paclitaxel resistant xenograft tumor growth in vivo.
