摘要:
目的:探讨谷氨酰胺对急性全脑缺血再灌注损伤时小肠黏膜Occludin蛋白表达的影响。方法:采用四血管阻断法建立大鼠全脑缺血再灌注动物模型。实验分4组,分别为假手术组、盐水对照组、奥扎格雷组和谷氨酰胺组,每组24只大鼠。建模成功后,盐水对照组立即腹腔注射2 mL生理盐水;奥扎格雷钠组按33.67 mg/kg腹腔注射奥扎格雷钠;谷氨酰胺组按33.67 mg/kg腹腔注射奥扎格雷钠和按0.4 g/kg腹腔注射体积分数20%的L丙氨酸L谷氨酰胺双肽注射液。再灌注6、12及24 h,每组各处死大鼠8只,取脑和小肠,光镜下观察脑组织海马区病理改变,免疫组化SP法测定小肠黏膜Occludin蛋白表达。结果:盐水对照组、奥扎格雷钠组和谷氨酰胺组于再灌注6、12和24 h海马锥体细胞结构紊乱、边界模糊,在12和24 h各时间点,4组大鼠肠黏膜Occludin蛋白表达差异有统计学意义(H=15.175和20.234,P均<0.001)。与假手术组比较,其余3组Ocdudin蛋白表达下降(P<0.05);与谷氨酰胺组相比,盐水对照组和奥扎格雷钠组表达下降(P<0.05)。结论:谷氨酰胺可以提高全脑缺血再灌注大鼠小肠组织中Occludin蛋白表达水平,从而减轻全脑缺血再灌注对小肠黏膜的损伤。
Abstract:
Aim:To study the impact of glutamine on the expression of Occludin in the intestinal mucosal tissue from the acute global cerebral ischemiareperfusion(GCIR) injury rats.Methods:The global brain ischemia rat model was established through the fourvessel occlusion method. The rats were divided into four groups: the sham operation group (control group), the saline group,the ozagrel group and the glutamine group. The latter 3 groups were established GCIR model.The saline group was given the same volume normal saline time after reperfusion;the ozagrel group was given the same volume normal saline in which ozagrel 33.67 mg/kg was dissolved at the same time;the glutamine group was given the same volume normal saline in which ozagrel 33.67 mg/kg and 20% glutamine 0.4 g/kg were dissolved respectively.The time of observation was 6,12,24 h after reperfusion.After that, every experimental animal was killed,and brain and intestine were obtained to investigate the pathological changes of the neurons in the hippocampus with the HE staining and the expression of Occludin in the intestinal mucosa with the immunohistochemistry.Results:At 6,12,24 h after reperfusion,the struceture of hippocampus cells were disordered and the bondaries of them were indistinct in the saline group,the ozagrel group and the glutamine group.The expression of Occludin protein has significant difference among the 4 gorups at 12,24 h(H=15.175,20.234,P<0.001).The expression of Occludin in the saline group, the ozagrel group and the glutamine group decreased than that in the sham operation group;the expression of Occludin in the glutamine group was higher than those in the saline group and the ozagrel group(P<0.05).Conclusion:Glutamine can protect the intestinal mucosal barrier in the acute global cerebral ischemiareperfusion injury in rats against the decrease of the expression of Occludin.
