摘要:
目的:探讨5氟尿嘧啶(5FU)对胰腺癌sw1990细胞增殖及相关基因表达的影响。方法:应用不同剂量5FU(0、50、100、200、400、800 mg/L)作用sw1990 24、48和72 h后收集细胞,MTT法检测细胞增殖抑制率,RTPCR检测p53、bax和pten mRNA的表达,Western blot法检测P53、BAX、 PTEN及pAKT蛋白的表达。结果:5FU抑制sw1990细胞的增殖,抑制作用呈现剂量依赖性(F剂量=11.024,P=0.012)。5FU作用后sw1990细胞p53、bax及pten mRNA表达均升高(F剂量=4.317,6.812和10.403,P均<0.05;F时间=3.058,0.906和0.451,P均>0.05);5FU作用后sw1990细胞P53及BAX蛋白相对表达量高于未处理组(t=9.025,24.562,P<0.05),pAKT蛋白相对表达量低于未处理组(t=7.239,P=0.042),PTEN蛋白表达量无明显变化(t=4.938,P=0.845)。结论:5FU可能通过调节sw1990中 p53、bax mRNA及P53、BAX、pAKT蛋白表达,抑制sw1990增殖。
Abstract:
Aim:To investigate the influence of 5fluorouracil(5FU) on the growth and expression of associated genes of pancreatic carcinoma cell sw1990.Methods:sw1990 was treated with different concentrations of 5FU(0,50,100,200,400 and 800 mg/L)for 24, 48,72 h, then was to detect the inhibition ratio by MTT method,and p53,bax,and pten mRNA by RTPCR and P53,BAX,PTEN, and phosphorylated AKT(pAKT) proteins by Western blot.Results:5FU could inhibit the proliferation of sw1990 in a concentration dependent manner(Fconcentration=11.024,P=0.012).5FU could increase the expression of p53,bax and pten mRNA in a concentration dependent manner(Fconcentration=4.317,6.812,and 10.403,P<0.05;Ftime=3.058,0.906,and 0.451,P>0.05).The P53 and BAX protein level in sw1990 cells treated with 5FU was higher and pAKT protein level was lower than those untreated(t=9.025,24.562,and 7.239,P<0.05),while the PTEN protein level did not change(t=4.938,P=0.845).Conclusion:5FU may inhibit proliferation of sw1990 cells by regulating the expression of p53,bax mRNA and P53,BAX and pAKT proteins.
