摘要:
 目的:比较不同剂量的放射治疗对人脑胶质瘤干细胞及其分化的胶质瘤细胞的杀灭作用,探讨胶质瘤干细胞与放疗耐受的相关性。方法:分离、培养并纯化人脑胶质瘤干细胞,并同时诱导纯化的肿瘤干细胞向胶质瘤细胞分化,对人脑胶质瘤干细胞及其分化后的胶质瘤细胞分别以0、2、3、5、8、12 Gy的6 MV X线照射后,继续培养72 h,采用MTT法检测细胞的增殖情况。结果:由胶质瘤干细胞分化出的子代胶质瘤细胞对放射治疗的耐受性下降,且随照射剂量增大,细胞生长抑制显著(F=125.971,P<0.001)。结论:脑胶质瘤干细胞分化后,对放射治疗的耐受性下降。
