摘要:
目的:探讨黄杞总黄酮对脑缺血再灌注大鼠的脑保护作用及可能机制。方法:48只SD大鼠随机分成6组,假手术组、模型组、黄杞总黄酮(200、100、50 mg/kg)组、脑心通(292 mg/kg)组,每组8只。采用线栓法建立脑缺血再灌注模型。进行神经行为评分和脑组织含水量测定。然后检测血清中一氧化氮(NO)水平、脑组织匀浆中丙二醛(MDA)含量、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSHPx)活性。结果:各组间神经行为评分、脑组织含水量、血清中NO水平、脑组织匀浆中MDA含量、SOD和GSHPx活性比较,差异均有统计学意义(F为35.305、40.259、15.338、21.022、10.497和12.381,P均<0.001)。除100 mg/kg黄杞总黄酮组GSHPx外,200和100 mg/kg黄杞总黄酮组各指标与模型组比较,差异均有统计学意义(P均<0.05)。结论:黄杞总黄酮对脑缺血再灌注损伤具有改善作用,可能与其抗氧化活性有关。
Abstract:
Aim: To investigate the protective effects of total flavonoids of engelhardtia roxburghiana(TFER) on brain of cerebral ischemic/reperfusion rats and the possible mechanism.Methods:A total of 48 SD rats were randomly divided into six groups (8 rats per group): sham group, model group, 200,100,50 mg/kg TFER groups and 292 mg/kg Naoxintong group.The model of cerebral ischemia/reperfusion was set up using suture method.The neurobehavioral scores and brain water content were determined,and then,serum NO level, MDA content, SOD and GSHPx activities were evaluated.Results:There were significant differences in the neurobehavioral scores, brain water content, NO level,MDA content,SOD and GSHPx activities among six groups(F=35.305, 40.259, 15.338, 21.022,10.497, 12.381,P<0.001).Except GSHPx of 100 mg/kg TFER group,there were significant differences in other indexes of 100 and 200 mg/kg TFER groups compared with those of model group(P<0.05).Conclusion:TFER possesses protective effects on brain of cerebral ischemic/reperfusion injury. The efficacy may be due to its antioxidant activity.
