摘要:
目的:观察吡咯烷二硫代氨基甲酸(PDTC)对食管癌细胞系EC1增殖、凋亡和细胞周期的影响。方法:用不同剂量PDTC(1、5、10、50、100 μmol/L)处理EC1细胞24、48和72 h后,MTT法观察细胞增殖情况,计算细胞增殖抑制率。用不同剂量PDTC(0、5、10、50、100 μmol/L)处理EC1细胞48 h后,测定细胞凋亡率和细胞周期。结果:PDTC可剂量依赖性和时间依赖性地抑制EC1细胞的增殖(F剂量=8.950,P=0.005;F时间=30.718,P<0.001;F交互=60.504, P<0.001)。不同剂量PDTC作用48 h后,随着PDTC剂量的增加,EC1细胞凋亡率增加(F=7.020,P=0.001),G0/G1期细胞比例降低(F=771.064, P<0.001),G2/M和S期细胞比例增加(F=963.002、309.332,P均<0.001)。结论:PDTC可以抑制食管癌细胞系EC1的增殖,诱导其发生凋亡,并将其阻滞在S期。
Abstract:
Aim:To investigate the effects of pyrrolidine dithiocarbamate(PDTC) on the proliferation, apoptosis and cell cycle in esophageal cancer EC1 cell line.Methods:Cell proliferation of EC1 treated by different dosage of PDTC(1,5,10,50,100 μmol/L) for 24, 48 and 72 h was oberserved by MTT.The apoptosis and cell cycles of EC1 treated by different dosage of PDTC(0,5,10,50,100 μmol/L) for 48 h were measured by flow cytometry. The cells without treatment were the control.Results:PDTC inhibited the proliferation of EC1 cells in dosedependent and timedependent manner(Fconcentration=8.950,P=0.005;Ftime=30.718,P<0.001;Finteraction=60.504, P<0.001).After the treatment of PDTC for 48 h, cell apoptosis rate of EC1 cells increased with the dosage of PDTC increasing(F=7.020,P=0.001),the percentage of cells in G0/G1 phase decreased(F=771.064, P<0.001),however, the percentages of cells in G2/M and S phase increased(F=963.002,309.332, P<0.001).Conclusion:PDTC could inhibit the proliferation of EC1 cells, induce EC1 apoptosis and arrest cell cycle in S phase in vitro.
