摘要:
目的:探讨细胞色素P450(CYP)1A1和X射线损伤修复的交叉互补基因(XRCC1)基因多态性与肺癌易感性的关系。方法:采用PCRRFLP方法检测100例肺癌患者和100例正常对照者CYP1A1、XRCC1多态基因型。结果:CYP1A1突变纯合基因型在肺癌组和对照组中的频率分别为11.0%和17.0%,差异无统计学意义(χ2=3.163,P>0.05)。XRCC1突变纯合基因型在肺癌组和对照组中的频率分别为44.0%和22.0%,差异有统计学意义(χ2=13.472,P<0.001),XRCC1突变纯合基因型携带者患肺癌的危险度增加[OR=3.316(95%CI 1.729~6.359)]。两基因联合分析,XRCC1与CYP1A1突变基因型联合携带者较单一基因型携带者患肺癌危险度增加[OR=6.243(95%CI 2.286~17.046)]。结论:XRCC1突变纯合基因型是肺癌的易感因素,CYP1A1与XRCC1突变基因型在肺癌发生中具有联合作用,可增加肺癌发生的危险性。
Abstract:
Aim:To study the association between genetic ploymorphism of cytochromp (CYP)1A1, Xray repair cross complementing group 1(XRCC1)and lung cancer.Methods:Using PCRRFLP to detect genetic ploymorphism of CYP1A1 and XRCC1 in 100 cases of lung cancer and 100 normal controls.Results:The frequencies of CYP1A1 mutational genotype in lung cancer group and control group were 11.0% and 17.0%, respectively,and there was no significant difference(χ2=3.163,P>0.05). The frequencies of XRCC1 mutational genotype were 44.0% and 22.0% in lung cancer group and control group, respectively,and there was significant difference(χ2=13.472,P<0.001). There was increased risk of lung cancer for individuals carrying mutational genotype of XRCC1 [OR=3.316(95%CI 1.729~6.359)]compared with subjects carrying wild genotype. Joint analysis of the two genes suggested that there was increased risk of lung cancer for individuals carrying both mutational genotype of XRCC1 and mutational genotype of CYP1A1 compared with subjects carrying single mutational genotype of CYP1A1 or XRCC1[OR=6.243(95%CI 2.286~17.046)].Conclusion:The mutational genotype of CYP1A1 and XRCC1 may contribute to the risk of developing lung cancer.
