摘要:
目的:观察五味草水提物与醇提物预处理对四氯化碳(CCl4)所致小鼠急性肝损伤的保护作用。方法:昆明种小鼠108只,随机分成9组,即正常组,模型组,益肝灵组,五味草醇提物和水提物低、中、高剂量组,均灌胃给药7 d,末次给药1 h后除正常组外,其他8组小鼠均腹腔注射体积分数0.2%的CCl4橄榄油溶液10 mL/kg,20 h后眼静脉采血,用以测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、山梨醇脱氢酶(SDH)活性,同时迅速剖取肝脏,观察肝组织形态变化,测肝组织中还原型谷胱甘肽(GSH)、丙二醛(MDA)及超氧化物歧化酶(SOD)水平。结果:模型组小鼠血清ALT、AST和SDH活性升高;肝组织中GSH和SOD水平降低,MDA水平升高。五味草水提物中、高剂量组及醇提物高剂量组肝组织变性明显减轻,以上指标较模型组有明显改善(P均<0.05)。结论:五味草水提物与醇提物预处理对CCl4所致小鼠急性肝损伤有保护作用。
Abstract:
Aim:To study the protective effect of water extract and ethanol extract of Corydalis taliensis Franch. pretreatment on acute liver injury induced by carbon tetrachloride(CCl4) in mice.Methods:A total of 108 Kunming mice were randomly allocated into 9 groups:normal group, model group, Yiganling group, low,medium,and high dose of ethanol extract of Corydalis taliensis Franch. groups(equivalent drug 1,2,4 g/kg), low,medium,and high dose of water extract of Corydalis taliensis Franch. groups (equivalent drug 1,2,4 g/kg).The mice in each group were lavaged for 7 d.1 h after the last administration, all mice except the normal group were injected with 0.2%(V/V) CCl4 at 10 mL/kg,after 20 h, the blood samples from the eye venous vein were obtained to determine ALT,AST,and SDH,at the same time,the left lobe of the liver was dissected to determine GSH,MDA and SOD.Results: CCl4 could induce liver injury, increase the levels of ALT,AST,SDH in serum,and the content of MDA in liver tissue,and decrease the contents of GSH and SOD in liver tissue,while in high dose of ethanol extract group and medium and high dose of water extract groups,the above parameters improved(P<0.05).Conclusion:The water and ethanol extracts of Corydalis taliensis Franch. have protective effect on acute liver injury induced by CCl4 in mice.
