摘要:
目的:观察AG490对前列腺癌DU145细胞增殖及STAT3信号通路相关蛋白表达的影响,探讨其抗癌机制。方法:DU145细胞分组培养,对照组单纯用DMEM高糖培养基培养,实验组给予终浓度为10、25、50及80 μmol/L 的AG490,分别培养24、48和72 h后,采用 MTT比色法测定细胞增殖抑制率,Western blot检测作用72 h后STAT3信号通路相关蛋白(pJAK2、STAT3、pSTAT3、Cyclin D1及BCLxL)。结果:AG490能够抑制DU145细胞的增殖,呈剂量(F=786.170,P<0.001)、时间依赖性(F=354.570,P<0.001)。AG490作用于DU145细胞72 h后, pJAK2、STAT3、pSTAT3、Cyclin D1及BCLxL蛋白的表达均降低。结论:AG490能抑制前列腺癌DU145细胞的增殖,其机制可能与阻断STAT3信号通路有关。
Abstract:
Aim:To observe the effect of AG490 on DU145 cells proliferation and expressions of STAT3 signaling pathways related proteins,and explore its anticancer mechanism.Methods:DU145 cells were grouped.The control group was cultured with DMEM high sugar culture medium,and the other groups were given AG490 at 10, 25, 50, and 80 μmol/L and cultured separately for 24,48 and 72 h.Using MTT method to detect the cell proliferation inhibition rate,and Western blot to test the expression levels of STAT3 signaling pathways related proteins(pJAK2, STAT3, pSTAT3, Cyclin D1 and BCLxL).Results:AG490 could significantly inhibit the DU145 cells proliferation in a dose(F=786.170,P<0.001) and timedependent manner(F=354.570,P<0.001).The protein expression levels of pJAK2, STAT3, pSTAT3, Cyclin D1 and BCLxL obviously reduced after 72 h AG490 treatment.Conclusion:AG490 can effectively inhibit cell proliferation of DU145 cells,which may be involved in its blocking STAT3 signaling pathways.
