摘要:
目的:探讨糖尿病(DM)结肠动力障碍大鼠远端结肠平滑肌细胞凋亡基因的表达。方法:雄性SD大鼠18只应用链脲佐菌素40 mg/kg尾静脉注射后成功建立DM结肠动力障碍大鼠模型,分为模型 6周和10周组;另18只分为正常对照6周和10周组,各组9只大鼠。应用实时荧光定量PCR检测各组大鼠远端结肠平滑肌细胞的bax、bcl2和caspase3 mRNA表达,采用免疫组化SP法检测其蛋白的表达。结果:不同组别和作用时间各组大鼠远端结肠平滑肌细胞bax、bcl2、caspase3 mRNA和蛋白的表达差异均有统计学意义(P均<0.001),模型组与同一时间点正常对照组以及模型10周组与6周组大鼠比较,远端结肠平滑肌细胞bax和 caspase3 mRNA和蛋白的表达增强, bcl2 mRNA和蛋白的表达降低(P<0.05)。结论:远端结肠平滑肌细胞凋亡基因bax、bcl2及caspase3表达的改变可能是DM大鼠结肠动力障碍发病机制之一。
Abstract:
Aim:To explore the gene expressions of apoptosis in distal colon smooth muscle cells in diabetes mellitus colon dysmotility rats.Methods:A total of 36 male SD rats were randomly divided into 4 groups which included normal control 6 weeks group,normal control 10 weeks group, model 6 weeks group and model 10 weeks group,9 rats in each group.Diabetes mellitus colon dysmotility was induced by streptozotocin(40 mg/kg).The expressions of bax,bcl2 and caspase3 mRNA in the distal colon smooth muscle cells were determined by realtime quantitative PCR. Immunohistochemistry SP method was used to detect the expressions of bcl2,bax and caspase2 proteins.Results:The expressions of bax and caspase3 mRNA and protein in model groups were higher than those in the normal control groups and the expressions of bcl2 mRNA and protein in model groups were lower than those in the normal control groups(P<0.05). The expressions of bax and caspase3 mRNA and protein in model 10 weeks group were higher than those in model 6 weeks group and the expressions of bcl2 mRNA and protein in model 10 week group were lower than those in model 6 week group(P<0.05).Conclusion:The expressions of bax,caspase3,and bcl2 in distal colon smooth muscle cell may contribute to the pathogenesis of diabetic colon dysmotility.
