摘要:
目的:探讨阿托伐他汀抑制高血压所致左心室肥厚(LVH)的作用。方法:24只自发性高血压大鼠(SHR)随机分为治疗和对照组,治疗组给予阿托伐他汀[30 mg/(kg?d)],对照组给予等体积的蒸馏水灌胃,10周后测定各组大鼠收缩压(pSB)、心率、左心室质量指数(LVWI)以及血清和心肌组织中血管紧张素Ⅱ(AngⅡ)、转化生长因子β1(TGFβ1)的水平。结果:治疗10周后,治疗组的LVWI、左室心肌及血液中AngⅡ和TGFβ1的含量明显低于对照组,差异均有统计学意义(t=9.798、3.221、2.602、3.483和3.394,P<0.05);治疗组LVWI与心肌组织中AngⅡ和TGFβ1的水平呈正相关(r=0.634、0.579,P=0.024、0.015)。结论:阿托伐他汀可以抑制自发性高血压大鼠心肌肥厚,其作用机制可能与其抑制血管紧张素Ⅱ和TGFβ1的表达有关。
Abstract:
Aim: To explore the inhibitory effect and mechanism of atorvastatin on left ventricular hypertrophy(LVH) in spontaneous hypertension rats(SHRs).Methods:Twentyfour SHRs were randomly divided into the treatment and control groups.The treatment group were treated with atorvastatin for 10 weeks at dose of 30 mg/(kg?d),and rats in the control group were treated with distilled water of the same volume.The SBP,the LVWI and the level of TGFβ1 and AngⅡ were determined by ELISA after ten weeks.Results:Compared with the treatment group,the LVWI,and the levels of TGFβ1 and AngⅡ in left ventricular tissue and blood in the control group were higher,and the differences were significant (t=9.798,3.221,2.602,3.483,and 3.394,P<0.05). The value of LVM1 was positively correlated with the level of AngⅡand TGFβ1 in left ventricular tissue(r=0.634 and 0.579,P=0.024 and 0.015).Conclusion: Atorvastatin can inhibit LVH of the SHRs, and the mechanism may be related to its inhibiting the expression of AngⅡand TGFβ1.
