摘要:
目的:观察山奈酚对鼻咽癌CNE2细胞周期分布及细胞周期素B1(CyclinB1)、细胞周期依赖性蛋白激酶1(Cdk1)表达的影响。方法:分别用0、20、40、60、80和100 μmol/L的山奈酚处理CNE2细胞。处理24、48和72 h后,应用MTT法测定CNE2细胞活力;处理24和48 h后用流式细胞术检测细胞周期;处理24 h后用RTPCR技术检测细胞CyclinB1及Cdk1 mRNA的表达水平。结果:随山奈酚作用剂量的增加和作用时间的延长,CNE2细胞活力逐渐降低(F浓度=385.194,F时间=237.324,F浓度×时间=13.757,P<0.001),细胞被阻滞于G2/M期(P<0.05);CNE2细胞中CyclinB1和Cdk1 mRNA的表达量随山奈酚作用浓度的增加而逐渐降低(F=95.682、154.871,P<0.001)。结论:山奈酚可能通过下调CNE2细胞CyclinB1和Cdk1 mRNA的表达水平,诱导G2/M期阻滞,抑制其增殖。
Abstract:
Aim:To study the effects of kaempferol on cell cycle status and CyclinB1,Cdk1 mRNA expressions in CNE2 cells.Methods:CNE2 cells were treated with 0,20,40,60,80,and 100 μmol/L kaempferol.24,48 and 72 h later,proliferation was determined by MTT assay;24 and 48 h later,cell cycle was detected by flow cytometry;24 h later,the expressions of CyclinB1 and Cdk1 mRNA were detected by RTPCR.Results:The CNE2 cell growth ability was inhibited by kaempferol in a time and dosedependent manner(Fdose=385.194,Ftime=237.324,Finteraction=13.757,P<0.001);CNE2 cells was blocked in G2/M phase (P<0.05);the expressions of CyclinB1 and Cdk1 mRNA decreased with the increase of kaempferol dose (F=95.682,154.871,P<0.001).Conclusion:Kaempferol can block CNE2 cells in G2 /M phase through decreasing the expressions of CyclinB1 and Cdk1 mRNA,and inhibit the cell proliferation.
