目的:检测白癜风患者外周血T淋巴细胞CD28、CTLA4的表达情况。方法:选择白癜风患者20例(进展期10例,稳定期10例)和健康对照10例,密度梯度离心法分离外周血单个核细胞(PBMC)后,应用流式细胞仪检测细胞表面CD28、CTLA4的表达。结果:3组外周血CD8+ T细胞CD28的表达差异无统计学意义(F=1.187,P=0.320),但外周血CD3+和CD4+ T细胞CD28和CTLA4的表达、以及CD8+ T细胞CTLA4的表达差异均有统计学意义(F=5.132、6.963、4.927、8.751和5.219,P均<0.05)。与对照组相比,进展期组CD3+和CD4+ T细胞CD28的表达及CD8+ T细胞CTLA4的表达差异无统计学意义(P>0.05),而CD3+和CD4+ T细胞CTLA4的表达降低(P<0.05);稳定期组CD3+和CD4+ T细胞CTLA4的表达与对照组比较,差异无统计学意义(P>0.05),但CD3+和CD4+ T细胞CD28的表达明显降低(P<0.05)。结论:白癜风患者外周血T淋巴细胞上CD28、CTLA4表达异常,可能与患者免疫功能紊乱及疾病发生发展有关。
Abstract:
Aim:To detect the expressions of CD28 and CTLA4 in peripheral blood T cells of patients with vitiligo.Methods:Twenty vitiligo patients including 10 cases of progressive stage and 10 cases of stationary stage,and 10 normal individuals served as controls were subjected to isolated peripheral blood mononuclear cells(PBMC) by density gradient centrifugation.The expressions of CD28 and CTLA4 in T cells were detected using flow cytometry.Results:There were significant differences in the expressions of CD28 and CTLA4 in CD3+ T cells,CD4+ T cells,and CTLA4 in CD8+ T cells among the three groups(F=5.132,6.963,4.927,8.751,and 5.219,P<0.05),but not in the expressions of CD28 in CD8+ T cells(F=1.187,P=0.320).Compared with those of the control group,the expressions of CD28 in CD3+ T cells and CD4+ T cells,and CTLA4 in CD8+ T cells in progressive stage vitiligo group were not significantly different(P>005),but those of CTLA4 in CD3+ T cells and CD4+ T cells were lower(P<0.05);the expressions of CTLA4 in CD3+ T cells and CD4+ T cells in stationary stage vitiligo group were not statistically different(P>0.05),but those of CD28 in CD3+ T cells and CD4+ T cells were lower(P<0.05).Conclusion:The abnormal expressions of CD28,CTLA4 in peripheral blood T cells in patients with vitiligo may induce functional disorders of costimulatory molecules,and may be relevant to the occurrence and development of vitiligo.
