摘要:
目的:观察二甲双胍对H2O2刺激的原代培养心肌细胞活性的影响。方法:利用乳鼠心室肌组织培养出大鼠原代心肌细胞,分别用不同浓度(0、1、10、100、1 000、3 000、5 000、10 000 μmol/L)的二甲双胍孵育0、15、30、60、120和180 min,然后再与100 μmol/L H2O2共孵育12 h,MTT法检测细胞活性。结果:当二甲双胍作用浓度从0增加到100 μmol/L,再增加到10 000 μmol/L时,H2O2刺激的心肌细胞活性先逐渐升高,后逐渐降低(F浓度=293536,P<0.001);随二甲双胍作用时间的延长,H2O2刺激的心肌细胞活性逐渐升高(F时间=185.438,P<0001)。进一步的观察结果显示,H2O2明显损伤原代心肌细胞的活性,100 μmol/L二甲双胍作用60 min可明显提高损伤细胞的活性(FH2O2=337.026,F二甲双胍=151.797,F交互=414.378,P均<0.001)。结论:低浓度二甲双胍对心肌细胞有明显的保护作用,可对抗活性氧对心肌细胞的损伤。
Abstract:
Aim:To assess the effects of metformin on cardiomyocytes damaged due to H2O2.Methods:Cardiomyocytes were pretreated with metformin at different doses(0,1,10,100,1 000,3 000,5 000,and 10 000 μmol/L)for different time(0,15,30,60,120 and 180 min),then were treated with 100 μmol/L H2O2 for 12 h.The viability of cells was analyzed by MTT method.Results:When pretreated with metformin at different doses,the viability of cardiomyocytes treated with H2O2 increased firstly,then decreased(Fdose=293.536,P<0.001);the viability of cardiomyocytes treated with H2O2 increased with the pretreatment time of metformin(Ftime=185.438,P<0.001).The further research results suggested that H2O2 could damage the primary cardiomyocytes,and 100 μmol/L metformin for 60 min could increase the viability of the cells (FH2O2=337026,Fmetformin=151.797,Finteraction=414.378,P<0.001).Conclusion:A low dose of metformin has beneficial effects on cardiomyocytes damaged by H2O2.
