摘要:
目的:研究肺间质纤维化大鼠肺组织中TGFβ1基因表达沉默对疾病进展的影响。方法:构建TGFβ1腺病毒siRNA载体。75只大鼠随机分为空白对照组15只,模型对照组、无关siRNA组和siRNA干预组各20只,后3组大鼠气管内滴入博莱霉素建立肺间质纤维化模型,24 h后,空白对照组、模型对照组分别经鼻滴入100 μL的生理盐水,无关siRNA组、siRNA干预组分别滴入等体积的重组腺病毒pAdenoXsiCon和pAdenoXsiTGF。第7、14、28天,采用免疫组化法检测肺组织TGFβ1蛋白的表达;第14 天,采用Realtime PCR和Western blot分别检测肺组织中Ⅰ、Ⅲ型胶原及TGFβ1 mRNA和蛋白的表达;第7、14、28天,采用ELISA法检测肺泡灌洗液和外周血中TGFβ1的表达水平。结果:第7、14、28天,4组大鼠肺组织中TGFβ1蛋白的表达和肺泡灌洗液中TGFβ1的水平差异均有统计学意义,第14天,4组大鼠Ⅰ、Ⅲ型胶原及TGFβ1 mRNA的表达差异亦有统计学意义(P均<0.05),且模型对照组、无关siRNA组和siRNA干预组以上指标均高于正常对照组,siRNA干预组以上指标低于模型对照组、无关siRNA组(P<0.05)。结论:沉默TGFβ1对大鼠肺间质纤维化的形成和发展可能有抑制作用。
Abstract:
Aim:To study the influence of silencing TGFβ1 gene expression in rats with pulmonary interstitial fibrosis(PIF) on the progress of the disease.Methods:The siRNA adenovirus vector was constructed and wrapped as adenovirus particle.A total of 75 rats were randomly allocated into blank control group(n=15),model control group(n=20),siRNAirrelevant group(n=20) and siRNA intervention group(n=20).The rat model with PIF was established in the latter 3 groups.After 24 h,the siRNAirrelevant group and siRNA intervention group were given pAdenoXsiCon and pAdenoXsiTGF,respectively.On the 7th,14th,and 28th day,the expression of TGFβ1 protein in lung tissue was detected by immunohistochemical method;on the 14th day,the mRNA and protein expressions of collagen Ⅰ and Ⅲ,and TGFβ1 in lung tissue were detected by Realtime PCR and Western blot;on the 7th,14th,and 28th day,the level of TGFβ1 in the bronchus alveolus lavage fluid and the peripheral blood was detected by ELISA.Results:On the 7th,14th,and 28th day,the expression of TGFβ1 protein in lung tissue and the level of TGFβ1 in the bronchus alveolus lavage fluid among the 4 groups had significant difference;on the 14th day,the expressions of collagen Ⅰ and Ⅲ,TGFβ1 mRNA in the lung tissue of the 4 groups also had significant difference(P<0.05),among which,the above parameters in the model control group,siRNAirrelevant group and siRNA intervention group were all higher than the blank control group,and those in the siRNA intervention group were lower than the model control group and siRNAirrelevant group(P<0.05).Conclusion:Silencing TGFβ1 may inhibit the formation and development of PIF in rats.
