摘要:
目的:探讨褪黑素对大鼠急性脊髓损伤后白细胞介素10(IL10)表达的影响,以及对损伤后炎症反应、神经修复的作用。方法:108只成年SD大鼠随机分为模型组、治疗组和对照组3组。前2组用改良的WD法建立T12脊髓损伤模型,对照组仅切除T12椎板不损伤脊髓。治疗组在脊髓损伤后10 min注射褪黑素(100 mg/kg),模型组注射等量的体积分数5%乙醇。术后第1、2、3、5、7天对3组大鼠进行神经功能评分,检测血清中IL10含量和脊髓中IL10 mRNA的水平。3 d时各组选6只行主动脉灌流,后取脊髓进行HE染色,免疫组化SP法染色观察IL10的表达特点。结果:模型组和治疗组大鼠术后均出现了严重的下肢神经功能障碍,BBB运动功能评分早期差异无统计学意义(P>0.05),术后3天、5天、7天两组运动功能评分差异有统计学意义(P<0.01)。术后同时间点治疗组脊髓内IL10 mRNA水平均高于模型组和对照组,这与血清中IL10含量及表达IL10阳性细胞数结果一致,均有治疗组最高、模型组次之、对照组最低,各组之间差异具有统计学意义(P<0.01);免疫组化见IL10在中央管室管膜细胞、神经细胞、神经胶质细胞胞浆中分布较多。结论:褪黑素可能通过提高中央管周围内皮细胞、神经细胞、胶质细胞对IL10的分泌来抑制脂质过氧化反应和炎症反应。
Abstract:
Aim: To explore the effects of melatonin (MT) on expression of interleukin10(IL10)in acute spinal cord injury (SCI) in rats, and to investigate the function of MT in the inflammatory response and nerve repair after acute SCI. Methods: A total of 108 SpragueDawley (SD) rats were randomly divided into 3 groups: model group, treatment group and control group. The first two groups acute SCI model were established by using the modified Allen's weight drop (WD) method at T12 level, and the control group was removed lamina only. At 10 min after the model establishment, 5% ethanol was given to the model group and 100 mg/kg of MT to treatment group. At 1 d, 2 d, 3 d, 5 d, 7 d, the function of the spinal cord was assessed by Basso Beattie Bresnahan locomotor rating scale (BBB scale),the blood samples and spinal cord were obtained to measure the levels of IL10 and IL10 mRNA. At 3 d, 6 samples of spinal cord from aorta perfusion model were taken in each group for histological examination using HE and Immunohistochemical SP staining methods. Results:Rats of model group and treatment group showed severe lower limb neurological dysfunction after surgery. At 3 d,5 d and 7 d, the difference between them in BBB scale had statistical difference (P<0.01). IL10 mRNA levels of treatment group was highest at each time point, and it was consistent with the IL10 levels at serum and the IL10 positive cells. The difference between each two groups had statistical significant (P<0.01), immunohistochemistry showed that more of IL10 distributed at the cytoplasm of endothelial cell, nerve cells and glial cell.Conclusion:This study indicates that MT suppress lipid oxidation reactions and inflammatory response by increasing the secretion of IL10 in central canal surrounding endothelial cell, nerve cells and glial cell.
