摘要:
目的:探讨重组人粒细胞集落刺激因子(rhGCSF)对急性缺血再灌注大鼠脑缺血半暗带区Survivin、Egr1表达和微血管再生的影响。方法: 72只大鼠随机分为假手术组、模型组和治疗组,每组24只。模型组和治疗组大鼠制备大脑中动脉闭塞(MCAO)再灌注局灶性脑缺血大鼠模型。治疗组大鼠术后2 h后自腹部皮下注射rhGCSF 50 μg/(kg·d),单次注射7.5 mL,连续用药5 d,模型组和假手术组给予等体积生理盐水。各组大鼠均于实验后7、14和21 d处死8只。免疫组化SP法检测Egr1和Survivin蛋白的表达,并通过CD31+细胞的标记进行微血管计数。结果:实验后7、14和21 d,各组大鼠脑组织缺血半暗带Egr1和Survivin蛋白表达的比较,差异有统计学意义(F=194.983、497.289、122.182、162.468、322.983和126.100,P<0.001)。实验后14 d,2种蛋白的表达达到峰值,之后逐渐下降。实验后14 d,治疗组Egr1和Survivin蛋白的表达均高于模型组。实验后14 d微血管计数达到高峰,治疗组高于模型组(P<0.05)。结论:rhGCSF可能通过上调Egr1和Survivin的表达促进血管形成。
Abstract:
Aim:To investigate the Survivin,early growth response1(Egr1) ,and vascular regeneration in ischemia half dark area of acute ischemia reperfusion rat mobilized by recombinant human granulocyte colony stimulating factor (rhGCSF).Method:A total of 72 SD rats were divided into 3 groups: the sham operation group, model group and treatment group,24 rats in each group. The rats in the model and treatment groups made MCAO model. The rats in the treatment group injected rhGCSF at 50 μg/(kg·d) 2 h after operation for 5 days, The model group and sham operation group were given normal saline. The expressions of Egr1 and Survivin proteins were detected by immunohistochemistry.The microvascular was marked by CD31 staining. Results: At 7,14,21 d after experiment the expressions of Egr1 and Survivin proteins among the 3 groups were obviously different(F=194.983,497.289,122.182,162.468,322.983 and 126.100,P<0.001).The expressions reached the peak at 14 d. The number of microvascular climbed to the top at the same time(P<0.05). At 14 d after experiment,the expressions of Egr1 and Survivin and the number of microvascular in the treatment group were higher than those in the model group.Conclusion: rhGCSF could promote the vascular regeneration through improving the expressions of Egr1 and Survivin proteins.
