摘要目的:观察p53抑制剂pifithrinα对肝原代细胞增殖及增殖相关蛋白表达的影响。方法:肝原代细胞分别经0、10、20和30 μmol/L的pifithrinα处理1 h后,再用10 nmol/L EGF诱导30、36和48 h,用放射性计数法测定细胞增殖水平。另取肝原代细胞,分3组,对照组常规培养,EGF组用10 nmol/L的EGF诱导,EGF+pifithrinα组用30 μmol/L pifithrinα和10 nmol/L EGF诱导,EGF诱导24、30 h后,Western blot法检测细胞中P53、P21、Cyclin D、Cyclin E、pErk蛋白的表达水平。结果:pifithrinα作用后,EGF诱导的细胞增殖水平均降低,且pifithrinα浓度越大,增殖水平越低(F剂量=54.690,F时间=214.370,F交互=50.450,P<0.001)。与对照组比较,EGF组细胞P53、Cyclin E蛋白表达水平未发生变化,但P21、Cyclin D及 pErk表达水平升高(P<0.05),EGF+pifithrinα组细胞P21、Cyclin D及pErk蛋白表达水平较EGF组降低(P<0.05)。结论:P53蛋白可能通过激活MAPK信号通路、上调Cyclin D表达,从而发挥促进肝原代细胞增殖的作用。
Abstract:
AbstractAim: To observe the effects of pifithrinα, the p53 inhibitor, on the proliferation and expression of the proliferationrelated proteins in primary hepatocytes.Methods: Primary hepatocytes were treated with 0,10,20 and 30 μmol/L pifithrinα for 1 h, stimulated by 10 nmol/L EGF for 30,36 and 48 h,then the proliferation was measured using method of thymidine incorporation. Primary hepatocytes were treated with 10 nmol/L EGF(EGF group),30 μmol/L pifithrinα and 10 nmol/L EGF(EGF+pifithrinα group) for 24 and 30 h,respectively, then the expressions of P53, P21, Cyclin D, Cyclin E and pErk proteins were checked by Western blot.The cells cultured regularly were the control.Results: Proliferation of the cells induced by EGF was inhibited by pifithrinα(Fdose=54.690,Ftime=214.370,Finteraction=50.450,P<0.001).The expressions of P21, Cyclin D and pErk in EGF group were higher than that in the control(P<0.05),the above parameters in EGF+pifithrinα group were lower than those in EGF group(P<0.05). Conclusion: P53 protein could induce proliferation of primary hepatocytes through activating MAPK pathway to upregulate the expression of Cyclin D.
