摘要目的:观察葛根素对阿尔茨海默病(AD)模型大鼠的治疗效果。方法: 成年SD雄性大鼠80只随机分成对照组、模型组、雌二醇组和葛根素组,每组20只。后3组采用D半乳糖联合亚硝酸钠制备AD模型。雌二醇组和葛根素组分别采用雌二醇和葛根素治疗8周。用水迷宫实验测各组大鼠学习记忆力的变化后,处死,测脑组织中脑蛋白含量、TchE活力、MDA含量、GSHPX活力、超氧阴离子自由基清除率及超氧阴离子自由基浓度。结果:与模型组比较,雌二醇组和葛根素组大鼠的学习记忆力明显改善(P<0.001),脑组织中TchE活力、MDA含量和超氧阴离子自由基浓度降低(F=22 171.583,517 912.892,55 461.776,P<0.001),脑蛋白含量、GSHPX活力及超氧阴离子自由基清除率升高(F=1 025.642,19 931.812,715 851.361,P<0.001);与雌二醇组比较,葛根素组各检测指标差异无统计学意义(P>0.05)。结论:葛根素对AD模型大鼠有治疗效果,且与雌二醇疗效相近。
Abstract:
Abstract Aim: To observe the therapeutic effect of puerarin on the rat model with Alzheimers disease (AD). Methods: Eighty adult male SD rats were randomly divided into control group, model group, estradiol treatment group and puerarin treatment group (n=20). The latter three groups were established rat AD model using the D galactose united sodium nitrite. The two treatment groups were given estradiol and puerarin treatment for 8 weeks. The water maze experiment was used to observe learning and memory changes of rats. And then the rats were put to death, the simultaneous detection of brain protein content,TchE, MDA content, GSHPX activity, superoxide anion radical scavenging and superoxide anion radical concentration in rat brain homogenate was performed. Results: Compared with model group, the learning and memory ability of the estradiol treatment group and puerarin treatment group improved significantly (P<0.001), the TchE, MDA content and superoxide anion radical concentration in the brain tissue of two treatment groups decreased (F=22 171.583,517 912.892,55 461.776, P<0.001),and brain protein content, superoxide anion radical scavenging and GSHPX activity increased(F=1 025.642,715 851.361,19 931.812, P<0.001). Compared with estradiol treatment group, the indexes in puerarin treatment group had no significant difference (P>0.05). Conclusion: Estrogen has therapeutic effect on the AD model rats. Moreover, puerarin and estradiol have similar therapeutic effect.
