摘要目的:检测卵巢上皮性癌组织中SFRP5基因启动子区甲基化与βcatenin蛋白的表达。方法:应用甲基化特异性PCR法和免疫组织化学SP法分别检测20例正常卵巢上皮组织、20例卵巢交界性上皮性肿瘤组织以及40例卵巢上皮性癌组织中SFRP5基因启动子区甲基化状态以及βcatenin蛋白的表达。结果:在正常卵巢上皮、卵巢交界性上皮性肿瘤以及卵巢上皮性癌组织中,SFRP5基因启动子区甲基化率分别为0.0%、10.0%、67.5%,差异有统计学意义(P<0.001);βcatenin蛋白的异常表达率分别为5.0%、40.0%、72.5%,差异有统计学意义(χ2=24.962,P<0.001)。卵巢上皮性癌组织中βcatenin蛋白的异常表达与SFRP5基因启动子区甲基化有关联(rP=0.468,P<0.001)。结论:SFRP5基因启动子区甲基化可能导致βcatenin蛋白异常表达,并可能通过激活Wnt/βcatenin信号传导通路促进卵巢上皮性癌的发生发展。
Abstract:
AbstractAim: To investigate the promoter region methylation of SFRP5 and its relationship with the expression of βcatenin protein in epithelial ovarian carcinoma tissue. Methods: Methylationspecific PCR and immunohistochemical SP method were used to separately detect the promoter region methylation status of SFRP5 and the expression of βcatenin protein in 20 cases of normal epithelial ovarian tissue, 20 cases of borderline epithelial ovarian tumor tissue and 40 cases of epithelial ovarian carcinoma tissue. Results: The methylation rate of SFRP5 was 0.0%,10.0%,and 67.5% in normal epithelial ovarian tissue, borderline epithelial ovarian tumor and epithelial ovarian carcinoma tissue(P<0.001);the abnormal expression rate of βcatenin protein was 5.0%,40.0%,and 72.5% in the above 3 groups(χ2=24.962,P<0.001). The abnormal expression of βcatenin protein was associated with the promoter region methylation of SFRP5(rP=0.468, P<0.001). Conclusion: The promoter region methylation of SFRP5 might cause the increasing expression of βcatenin protein, which might activate Wnt/βcatenin signaling pathway and contribute to the carcinogenesis and development of epithelial ovarian carcinoma.
