摘要目的:探讨抑制肝癌SMMC7721细胞中TWIST基因表达对其侵袭能力的影响。方法:将靶向TWIST基因的siRNA序列和阴性对照siRNA序列转染入人肝癌SMMC7721 细胞,同时以未转染细胞作空白对照。于转染后48 h,采用半定量RTPCR和Western blot法检测各组细胞TWIST、Ecadherin mRNA和蛋白的表达,Boyden侵袭小室检测细胞体外侵袭能力的变化。结果:3组TWIST、Ecadherin mRNA和蛋白表达,以及穿膜细胞数差异有统计学意义(F=32.023、38.732和24.034、49.083、44.387,P<0.001);TWIST siRNA组TWIST mRNA和蛋白的表达低于空白和阴性对照组,Ecadherin mRNA和蛋白的表达高于空白和阴性对照组,TWIST siRNA组穿膜细胞数较空白对照组和阴性对照组下降(P<0.05)。结论:抑制肝癌SMMC7721细胞TWIST基因表达可以抑制肝癌细胞上皮间质转化的发生,同时可有效降低肝癌细胞的侵袭转移能力。
Abstract:
AbstractAim: To explore the effect of silencing TWIST by siRNA on the invasion capability of SMMC7721 cell line. Methods:siRNA targeting TWIST and siRNA as a negative control were chemically synthesized, and were transfected into SMMC7721 cells. SMMC7721 cells without transfection were used as blank control.The levels of TWIST and Ecadherin mRNA and protein were evaluated by semiquantitative RTPCR and Western blot after 48 h. Boyden chamber was used to evaluate the invasion capability of SMMC7721 cells in vitro. Results: The levels of TWIST and Ecadherin mRNA and protein,and the number of cells under micromembrane among the 3 groups were significantly different(F=32.023,38.732,24.034,49.083,44.387,P<0.001). The levels of TWIST mRNA and protein in TWIST siRNA group were lower than those in blank and negative control groups. The levels of Ecadherin mRNA and protein in TWIST siRNA group were higher than those in blank and negative control groups. The number of cells under micromembrane decreased in TWIST siRNA group compared with those in blank and negative control groups(P<0.05).Conclusion: Inhibiting the expression of TWIST gene in SMMC7721 cells could inhibit the epithelial mesenchymal transition and can reduce their invasion capability.
